Functional Difference of MYC and MYCN in Combined Hepatocellular-Cholangiocarcinoma: Regulation of Differentiation by HNF1B. [PDF]

open access: yesCancer Sci
Combined hepatocellular–cholangiocarcinoma (cHCC‐CCA) is a tumor consisting of a mixture of hepatocellular carcinoma and intrahepatic cholangiocarcinoma (CCA) components, but the mechanisms of development are poorly understood. The activation of Myc may be implicated in the formation of cHCC‐CCA and this work demonstrates for the first time that Myc ...

Hepatocyte BPGM Induces RET Lactylation and Macrophage Reprogramming to Promote Tumorigenesis in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
Mechanistic schematic diagram.BPGM promotes the expression level of RET by increasing the lactylation of RET‐K549 and inhibiting its ubiquitination levels in HCC cells. Furthermore, BPGM in HCC cells could also promote M2 polarization in macrophages through lactate secretion. Both of the mechanisms could promote the progression of HCC. ABSTRACT Aerobic

Inferring Gene Regulatory Networks From Single‐Cell RNA Sequencing Data by Dual‐Role Graph Contrastive Learning

open access: yesAdvanced Science, EarlyView.
RegGAIN is a novel and powerful deep learning framework for inferring gene regulatory networks (GRNs) from single‐cell RNA sequencing data. By integrating self‐supervised contrastive learning with dual‐role gene representations, it consistently outperforms existing methods in both accuracy and robustness.

TAF2: A potential oncogene for hepatocellular carcinoma [PDF]

open access: yes, 2017
Astrocyte Elevated Gene 1 (AEG1) is an oncogene for hepatocellular carcinoma (HCC). Its role in HCC pathogenesis has been well studied. A pan cancer analysis of gene expression in multiple databases identified TATA-box binding protein associated factor 2

SIRT5–RAC2 Axis Drives Monocyte‐to‐Macrophage Differentiation to Promote Inflammatory Injury in Premature Ovarian Insufficiency

open access: yesAdvanced Science, EarlyView.
SIRT5 desuccinylates and stabilizes RAC2, activating CSF1R‐dependent signaling to drive monocyte differentiation into M0 macrophages and their polarization toward pro‐inflammatory M1 phenotypes in CTX‐induced premature ovarian insufficiency. Inhibiting the SIRT5‐RAC2 axis attenuates inflammation, reduces granulosa cell apoptosis, and preserves ...

Chaperone‐Mediated Autophagic Degradation of USP9X in Macrophages Exacerbates Postmyocardial Infarction Inflammation and Cardiac Dysfunction

open access: yesAdvanced Science, EarlyView.
This study demonstrates that inflammatory stimuli induce the acetylation‐triggered, chaperone‐mediated autophagic degradation of ubiquitin‐specific peptidase 9 X‐linked (USP9X) in macrophages. USP9X acts as a macrophage “inflammation switch” after myocardial infarction (MI). USP9X loss destabilizes tumor necrosis factor receptor‐associated factor (TRAF)

Expression of the oncogenes mil and ras abolishes the in vivo differentiation of mammary epithelial cells [PDF]

open access: yes, 2017
Three carcinoma-associated oncogenes, two of which have been strongly implicated in human mammary tumorigenesis, have been introduced into a novel mouse mammary epithelial cell line, EF43, that retains many differentiated functions.

Targeting PRKCN, an Essential Driver Orchestrating mTOR‐IRF4 Axis Independently of Kinase Activity, in Multiple Myeloma

open access: yesAdvanced Science, EarlyView.
Constitutive PRKCN expression is driven by super‐enhancers and modulated by NF‐κB signaling in multiple myeloma (MM). PRKCN activates mTORC1/2‐IRF4 signaling axis and favors tumor cell growth independently of its kinase activity. IRF4 reciprocally promotes PRKCN transcription, creating a feed‐forward loop.

The NF-κB subunit c-Rel regulates Bach2 tumour suppressor expression in B-cell lymphoma

open access: yes, 2016
The REL gene, encoding the NF-κB subunit c-Rel, is frequently amplified in B-cell lymphoma and functions as a tumour-promoting transcription factor. Here we report the surprising result that c-rel–/– mice display significantly earlier lymphomagenesis in ...

Gut Microbiota‐Non‐Coding RNA Axis in Immune Modulation and Disease: From Mechanisms to Clinical Translation

open access: yesAdvanced Science, EarlyView.
ABSTRACT Immune homeostasis is indispensable for preserving organismal integrity, orchestrated through complex molecular networks encompassing immune cell dynamics, microbial cues, and epigenetic regulation. Among these, the gut microbiota‐non‐coding RNA (ncRNA) axis has recently garnered substantial attention as a multifaceted modulator of host ...
