The Intersection of Purine and Mitochondrial Metabolism in Cancer
Nucleotides are essential to cell growth and survival, providing cells with building blocks for DNA and RNA, energy carriers, and cofactors. Mitochondria have a critical role in the production of intracellular ATP and participate in the generation of ...

This study investigates hypoglycemia‐induced diabetic macrovascular endothelial dysfunction. It reveals that hypoglycemia triggers ZBP1‐dependent PANoptosis of endothelial cells, proinflammatory polarization of macrophages, and fibrosis of vascular smooth muscle cells (VSMCs) in diabetic mice.

During atherosclerosis, endothelial cells release purines in response to oxidized phospholipids. Here, Hitzel et al. show that oxidized phospholipids activate an MTHFD2-regulated gene network in endothelial cells which reprograms amino acid metabolism ...

Substituted Purines as High-Affinity Histamine H3 Receptor Ligands
Continuing with our program to obtain new histamine H3 receptor (H3R) ligands, in this work we present the synthesis, H3R affinity and in silico studies of a series of eight new synthetically accessible purine derivatives.

The Influence of Dietary Purines and Pyrimidines on Purine and Pyrimidine Biosynthesis in Man
Allopurinol-induced orotaciduria is reduced by dietary ribonucleic acid (RNA), RNA hydrolysate and different nucleotides. These findings are compatible with feedback regulation of pyrimidine biosynthesis by dietary nucleotides. Serum uric acid and urinary uric acid excretion on a purine-free isoenergetic diet reach a minimum after about 10 days and ...

In SETD2‐mutant ccRCC, the original LEDGF recognition of the H3K36me3 regulatory axis no longer exists. LEDGF interacts with CARM1‐dependent H3R17me2a regulating the transcription of key enzymes in the de novo synthesis pathway. The abnormally elevated LEDGF leads to the accumulation of purine nucleotides in SETD2‐mutant ccRCC, thereby satisfying the ...

Reconstructive Methodology in the Synthesis of 2-Aminopurine
A fundamentally new synthetic approach to the synthesis of 2-aminopurine has been developed. It consists in the combination of the creation of a condensed polyazotic heterocyclic tetrazolopyrimidine structure, its transformation into triaminopyrimidine ...

This study uncovers CHKA as a pivotal driver of vascular dysfunction in diabetic retinopathy and highlights its role in endothelial dysfunction through NAD⁺‐SIRT1‐Notch signaling. Single‐cell and functional analyses, combined with clinical and genetic validation, suggest CHKA as a promising therapeutic target for the prevention and treatment of ...
