Interferon induction and antiviral activity was discovered with 2-amino-5-bromo-6-phenyl-4(3H)-pyrimidinone. An analogue study incorporating a series of 2-amino-5-substituted-6-arylpyrimidinones revealed that the most potent interferon inducers were mono- and difluorophenyl analogues.

Conformation of 1,2,3,4-tetrahydro-2-pyrimidinones

The conformation of a number of 1,2,3,4-tetrahydro-2-pyrimidinones with various substituents in the ring was established on the basis of the vicinal 3J34 constants found from the 1H NMR spectra of these compounds. It is shown that in solution rapid conformational transformations between the two possible “boat” conformations evidently occur.

Regioselectivity in the reactions of aryltri-isopropoxytitanium with pyrimidinones.

Complete regioselectivity is observed in the 1 : 1-adduct formation between aryltri-isopropoxytitanium reagents and pyrimidin-2(1H)ones; the new carbon–carbon bond is formed at C-4. Dehydrogenation gives the arylated, fully conjugated heterocycle.

Microwave‐assisted copper powder‐catalyzed coupling and cyclization of β‐bromo‐α,β‐unsaturated amides with amidine hydrochlorides leading to pyrimidinones

β-Bromo-α,β-unsaturated amides are coupled and cyclized with amidine hydrochlorides using microwave irradiation in the presence of a catalytic amount of copper powder and a base to give the corresponding pyrimidinones in good yields.
