Isolation and sequence analysis of the cDNA encoding Δ1-pyrroline-5-carboxylate reductase from Zalerion arboricola

Gene, 1996
A cDNA encoding delta 1-pyrroline-5-carboxylate reductase (P5CR) was isolated from the pneumocandin (Pmo)-producing fungus, Zalerion arboricola (Za), by complementation of a P5CR-deficient mutant (pro3) of Saccharomyces cerevisiae (Sc). The cloned cDNA was placed under control of the Sc galactokinase (GAL1) promoter and restored P5CR activity to the ...

Δ1-pyrroline-5-carboxylate reductase from baker's yeast. Further purification by affinity chromatography with 5′ AMP-sepharose 4B

Biochimica Et Biophysica Acta - Biomembranes, 1980
A homogeneous preparation of delta 1-pyrroline-5-carboxylate reductase (L-proline: NAD (P)+ 5-oxidoreductase, EC 1.5.1.2) was obtained from baker's yeast by an affinity chromatography, using 5' AMP-Sepharose 4B. After the 1st DEAE-Sephadex column chromatography, the enzyme absorbed on 5' AMP-Sepharose column was eluted with 1 mM ATP.

Disease of Ornithine-Proline Pathway: A △1-pyrroline-5-carboxylate Reductase Deficiency in the Retina of Retinal Degeneration Mice

1982
The discovery of a genetic defect in ornithine oxoacid aminotransferase (L-ornithine: 2-oxoacid aminotransferase, EC 2.6.1.13) in gyrate atrophy of the choroid and retina1,2,3, prompted us to study the ornithine metabolism of the eye4. We soon found high 361 activities of ornithine oxoacid aminotransferase and △1-pyrroline-5-carboxylate reductase (L ...

Human pyrroline‐5‐carboxylate reductase (PYCR1) acts on Δ1‐piperideine‐6‐carboxylate generating L‐pipecolic acid

Journal of Inherited Metabolic Disease, 2014
AbstractWe have conducted biochemical studies with commercial available pyrroline‐5‐carboxylate (P5C) reductase (PYCR1) to investigate whether this enzyme plays a role in L‐lysine degradation. Our recent studies with antiquitin/ALDH7A1 deficient fibroblasts revealed an alternative genesis of L‐pipecolic acid, and we then hypothesized that PYCR1 was ...

delta 1-Pyrroline-5-carboxylate reductase from Baker's yeast. Purification, properties and its application in the assays of L-delta 1-pyrroline-5-carboxylate and L-ornithine in tissue.

Biochimica et biophysica acta, 1981
delta 1-Pyrroline-5-carboxylate reductase (L-proline:NAD(P)+ 5-oxidoreductase, EC 1.5.1.2) from Baker's yeast has been purified and characterized. Purification to an apparently homogenous protein was effected by using 'reagent-grade' water containing dithiothreitol and by maintaining a constant pH 7.5, because of the instability of the enzyme protein ...

A soybean gene encoding Δ1-pyrroline-5-carboxylate reductase was isolated by functional complementation in Escherichia coli and is found to be osmoregulated

Molecular and General Genetics MGG, 1990
We have isolated several cDNA clones encoding delta 1-pyrroline-5-carboxylate reductase (P5CR, L-proline: NAD(P)+ 5-oxidoreductase, EC 1.5.1.2) which catalyzes the terminal step in proline biosynthesis, by direct complementation of a proC mutation in Escherichia coli with an expression library of soybean root nodule cDNA. The library was constructed in

Integrative bioinformatics and experimental validation identify pyrroline-5-carboxylate reductase 1 as a mitochondrial regulator and diagnostic biomarker in osteosarcoma

International Journal of Biological Macromolecules
Osteosarcoma (OS) is a highly aggressive bone malignancy with limited effective biomarkers and therapeutic strategies. An integrative analysis was conducted combining transcriptomic profiling, mitochondrial-related gene (MRG) screening, and machine learning approaches to uncover novel diagnostic and prognostic markers.
