Erweiterung des Repertoires an lichtschaltbaren unnatürlichen Aminosäuren für das Enzym‐Engineering

open access: yesAngewandte Chemie, Volume 137, Issue 38, September 15, 2025.

Expanding the Repertoire of Photoswitchable Unnatural Amino Acids for Enzyme Engineering

open access: yesAngewandte Chemie International Edition, Volume 64, Issue 38, September 15, 2025.
Enzyme engineering with photoswitchable unnatural amino acids (psUAAs) has a high potential for applications such as biocatalysis. Here, we extend the current repertoire of psUAAs to include more versatile properties paving the way to more advanced photocontrol engineering.

Deciphering context-dependent amber suppression efficiency in mammalian cells with an expanded genetic code [PDF]

open access: yes, 2021
The genetic code of organisms can be expanded by introducing orthogonal translation systems (OTSs). One of the most commonly applied OTSs in mammalian cells is the archaeal pyrrolysyl-tRNA synthetase/tRNA_Pyl_CUA (PylRS/PylT) pair from Methanosarcina ...

Engineering a promiscuous pyrrolysyl-tRNA synthetase by a high throughput FACS screen [PDF]

open access: yes, 2017
AbstractThe Pyrrolysyl-tRNA synthetase (PylRS) and its cognate tRNAPyl are used to facilitate the incorporation of non-canonical amino acids (ncAAs) into the genetic code of bacterial and eukaryotic cells by orthogonally reassigning the amber codon. Currently, the incorporation of new ncAAs requires a cumbersome engineering process composed of several ...

Site-Specific Incorporation of Selenocysteine by Genetic Encoding as a Photocaged Unnatural Amino Acid [PDF]

open access: yes, 2018
Selenocysteine (Sec) is a naturally occurring amino acid that is also referred to as the 21st amino acid. Site-specific incorporation of Sec into proteins is attractive, because the reactivity of a selenol group exceeds that of a thiol group and thus ...

Functional consequences of lysine acetylation of phosphofructokinase isozymes

open access: yesThe FEBS Journal, Volume 292, Issue 10, Page 2545-2558, May 2025.
Here, we investigated the impact of lysine acetylation on phosphofructokinase (Pfk) isozyme activity using a genetic code expansion strategy. We found that the acetylation of lysine residues in Pfk isozymes in Escherichia coli affects the activities of these enzymes and is generated by non‐enzymatic acetylation with acetyl phosphate.

MINFLUX nanoscopy: Visualising biological matter at the nanoscale level

open access: yesJournal of Microscopy, Volume 298, Issue 2, Page 219-231, May 2025.
Abstract Since its introduction in 2017, MINFLUX nanoscopy has shown that it can visualise fluorescent molecules with an exceptional localisation precision of a few nanometres. In this overview, we provide a brief insight into technical implementations, fluorescent marker developments and biological studies that have been conducted in connection with ...

Optimized Directed Evolution of E. coli leucyl‐tRNA Synthetase adds many Noncanonical Amino Acids into the Eukaryotic Genetic Code Including Ornithine and Nϵ‐Acetyl‐Methyllysine

open access: yesAngewandte Chemie, Volume 137, Issue 14, April 1, 2025.
The pyrrolysyl pair has dominated noncanonical amino acids (ncAAs) incorporation in eukaryotes, but additional engineerable pairs are needed to further expand this toolbox. Although E. coli leucyl‐tRNA synthetase (EcLeuRS) is a promising candidate, engineering its substrate specificity has been much less successful.

Incorporation of Multiple β2-Hydroxy Acids into a Protein In Vivo Using an Orthogonal Aminoacyl-tRNA Synthetase. [PDF]

open access: yes
The programmed synthesis of sequence-defined biomaterials whose monomer backbones diverge from those of canonical α-amino acids represents the next frontier in protein and biomaterial evolution.

Conformational transition of FGFR kinase activation revealed by site-specific unnatural amino acid reporter and single molecule FRET [PDF]

open access: yes, 2016
Protein kinases share significant structural similarity; however, structural features alone are insufficient to explain their diverse functions. Thus, bridging the gap between static structure and function requires a more detailed understanding of their ...
