Design, Synthesis, and Selective Antiproliferative Activity of Indolizine Derivatives as Microtubule Destabilizers

open access: yesArchiv der Pharmazie, Volume 358, Issue 12, December 2025.
A series of indolizine derivatives was synthesized and biologically evaluated. Compounds 8e and 8h showed nanomolar antiproliferative activity against cancer cells and low toxicity to normal cells. They act as microtubule‐destabilizing agents, inducing G2/M arrest and modulating key proteins. Docking confirmed binding at the colchicine site of tubulin.
