Genetically‐Programmed Hypervesiculation of Lactiplantibacillus Plantarum Increases Production of Bacterial Extracellular Vesicles with Therapeutic Efficacy in a Preclinical Inflammatory Bowel Disease Model
Advanced Science, EarlyView. The development of a novel engineered strain of Lactiplantibacillus plantarum (Lp) is reported that can produce bacterial extracellular vesicles (BEVs) at >60‐fold higher yields than the unmodified parental strain. These Lp BEVs retain therapeutic bioactivity as validated in a mouse model of dextran sodium sulfate‐induced colitis.Nicholas H Pirolli, Daniel Levy, Alyssa Schledwitz, Natalia Sampaio Moura, Mitali Sarkar, Talia J. Advanced Science, EarlyView.Amid the ongoing threat of emerging viral pathogens, host‐directed antivirals offer a strategy to overcome viral mutation and drug resistance. SB2960, a small‐molecule inducer of stress granules (SGs), exhibits potent broad‐spectrum antiviral activity with minimal cytotoxicity.Wan Gi Byun, Sumin Son, JinAh Lee, Minha Lee, Meehyun Ko, Songrui Zhao, Ju Hee Kim, Kannan Vaithegi, Ji Hoon Kwon, Jung Ho Lee, Sangeun Jeon, Dawon Yi, Peng Zou, Seungtaek Kim, Seung Bum Park +14 morewiley +1 more sourceWDR5‐H3K4me3 Epigenetic Axis Promotes TRMT6‐Dependent tRNA M1A Modification to Facilitate Triple‐Negative Breast Cancer Progression by Suppressing Ferroptosis
Advanced Science, EarlyView.Upregulated TRMT6 forms aberrant hypermethylation of a specific tRNA pool and serves as a predictor of poor prognosis in TNBC. This m1A modification in tRNAs enhances translation of FTH1 and FTL, reducing the pool of bioavailable Fe2⁺. Reduced Fe2+ availability impairs RSL3‐induced lipid peroxidation and tumor progression.Yuqing Lei, Xue Wang, Jiahe Liu, Zixu Niu, Jiaqi Hua, Qian Guo, Dan Du, Yuanchang Zhu, Fucheng He, Mingxia Zhou, Hongle Li, Jing He, Jun Li +12 morewiley +1 more sourceGupta K, Pearce S, Poursaid A, Aliyar H, Tresco P, Mitchnick M, Kiser P. 2008. Advanced Science, EarlyView.Extracellular vesicles (EVs) released from TGF‐β‐activated CAFs are enriched with ECM proteins such as TSG6 and THBS1, which facilitate their binding to recipient cell membranes. This EV–cell interaction promotes the clustering of CD44 and TGF‐β receptors on the target cell surface, thereby potentiating TGF‐β signaling activity. Advanced Science, EarlyView. In wild‐type intestinal epithelial cells, KDM6B demethylates SLC10A2 to promote its expression, after which SLC10A2 suppresses ERK phosphorylation, inhibiting AP‐1 (Fos/Jun) transcription and chemokine production. In KDM6B‐deficient intestinal epithelial cells, SLC10A2 methylation increases, and its expression decreases.Zhibo Hu, Jing Xun, Bin Liu, Xiaolin Jiang, Yingdi Han, Huichao Yang, Qi Gao, Ruifang Gao, Aimin Zhang, Xueliang Wu, Dapeng Zhang, Dihua Li, Tian Li, Shimin Yang, Ximo Wang, Xiangyang Yu, Qi Zhang +16 morewiley +1 more source
