BIOGENESIS of bacterial ribosomes proceeds through the formation of precursor particles1–4, consisting of an incomplete set of ribosomal protein5 and of RNA molecules that are longer and less methylated than mature RNA4,6,7. RNA maturation is commonly considered to be a part of the ribosome maturation process.

THE “rapidly labelled” RNA of immature erythroblasts includes a rather homogeneous high specific activity RNA in the 9S region of linear sucrose gradients, but when the same RNA is assayed for ability to stimulate protein synthesis in a cell-free system, the peak of activity is found just trailing the 18S ribosomal RNA1,2.

The analysis of RNA from L5178 Y cells, by electrophoreis in 1.7% polyacrylamide gel, after short time labelling with [methyl-3H] methionine, shows that the three molecular species 47S, 46S and 45S RNA from the ribosomal RNA "45S fraction" are methylated.

Splicing messenger RNA precursors: branch sites and lariat RNAs

Abstract The splicing of mRNA precursors proceeds through an unusual RNA structure: a lariat or tailed circular molecule where the 5′ end of the intervening sequence is attached to an internal residue via a 2′–5′ phosphodiester bond. The role of lariat formation and its possible significance are discussed.

Asymmetric maturation of a dimeric transfer RNA precursor

Abstract Six of the eight transfer RNAs coded by bacteriophage T4 are synthesized via three dimeric precursor molecules. The sequences of two of these have been determined. Both of these precursors give rise to equimolar amounts of the cognate tRNA molecules in vivo .

Isolation of ribosomal RNA precursors from Physarum polycephalum

Ribosomal RNA synthesis in Physarum polycephalum was studied by labeling intact microplasmodia with [3H]uridine. Labeled, high-molecular-weight RNA species were found in a 30,000 S structure released by phenol extraction at room temperature. RNA was released from the structure by further phenol extraction at 65–70 °C.
