Microfluidic Modeling of Macrophage‐Induced Cardiac Inflammation Using NF‐κB Reporter Cardiomyocytes
A cardiac inflammation model was developed to enable real‐time analysis of macrophage‐cardiomyocyte interactions using NF‐κB‐reporter cardiomyocytes and a microfluidic co‐culture system. A MATLAB‐based model was implemented to optimize cell density and medium resupply based on TNFα diffusion kinetics. Activation thresholds and stress responses in NF‐κB‐

Quantitative secretome analysis establishes the cell type-resolved mouse brain secretome [PDF]
AbstractTo understand how cells communicate in the nervous system, it is essential to define their secretome, which is challenging for primary cells because of large cell numbers being required. Here, we miniaturized secretome analysis by developing the high-performance secretome-protein-enrichment-with-click-sugars method (hiSPECS). To demonstrate its

A Synovium‐on‐Chip Platform to Study Multicellular Interactions in Arthritis
The Synovium‐on‐Chip comprises a thin microporous PDMS membrane to support co‐culture of fibroblast‐like synoviocytes (FLS), THP‐1‐derived macrophages, and endothelial cells, enabling real‐time analysis of synovial‐vascular interactions. FLS migration through the pores drives endothelial remodeling, while TNF‐α stimulation induces robust inflammatory ...

Secret3D Workflow for Secretome Analysis
Secretome analysis is crucial to unravel extracellular processes. However, secreted proteins are difficult to detect in mass-spectrometry-based analysis due to contamination of serum proteins deriving from cell culture media and to high glycosylation, which hampers tryptic digestion. Secret3D workflow is an optimized protocol for the global analysis of

Identification and analysis of seven effector protein families with different adaptive and evolutionary histories in plant-associated members of the Xanthomonadaceae. [PDF]
The Xanthomonadaceae family consists of species of non-pathogenic and pathogenic γ-proteobacteria that infect different hosts, including humans and plants.

A superhydrophobic 3D cell culture platform enables rapid and consistent formation of heterotypic tumor–stroma clusters, and reveals how physiological shear conditions influence metastatic signaling. Mechanical stimulation of cancer‐associated fibroblasts promotes sustained cytokine secretion and survival‐promoting pathways, showing how multicellular ...

Hepatocyte growth factor, a key tumor-promoting factor in the tumor microenvironment [PDF]
The tumor microenvironment plays a key role in tumor development and progression. Stromal cells secrete growth factors, cytokines and extracellular matrix proteins which promote growth, survival and metastatic spread of cancer cells.

3D Soft Hydrogels Induce Human Mesenchymal Stem Cells “Deep” Quiescence
Three‐dimensional soft hydrogels mimicking the bone marrow niche induce deep quiescence in human mesenchymal stem cells. Unlike 2D culture, 3D matrices halt proliferation, regulate cell‐cycle and quiescence markers, and downregulate mTORC1 signaling, preserving stem cell phenotype and therapeutic potential ex vivo.

Argonaute2 Regulates the Pancreatic β-Cell Secretome [PDF]
Argonaute2 (Ago2) is an established component of the microRNA-induced silencing complex. Similar to miR-375 loss-of-function studies, inhibition of Ago2 in the pancreatic β-cell resulted in enhanced insulin release underlining the relationship between these two genes.
