PRMT5‐Dependent Stabilization of VPS34 Orchestrates Copper Trafficking to Shield Cancer Cells from Cuproptosis and Radiotherapy

open access: yesAdvanced Science, EarlyView.
Radioresistance arises partly from tumor cells evading cuproptosis via unknown defenses. This study reveals that the PRMT5‐VPS34 axis acts as a radiation‐activated anticuproptotic mechanism: PRMT5 methylates VPS34 at Arg174, recruiting USP10 to remove K48‐linked ubiquitination and prevent degradation.

TDRD3 functions as a selective autophagy receptor with dual roles in autophagy and modulation of stress granule stability

open access: yes
Abstract Tudor Domain Containing 3 (TDRD3) is a methylarginine-reader protein that functions as a scaffold in the nucleus facilitating transcription, however TDRD3 is also recruited to stress granules (SGs) during the Integrated Stress Response (ISR) although its function therein remains largely unknown.

NOD1 deficiency promotes inflammation via autophagic degradation of ASK1

open access: yesCommunications Biology
Nucleotide-binding oligomerization domain-containing protein 1 (NOD1) is a pattern recognition receptor of bacterial peptidoglycans. NOD1 facilitates the elimination of invading intracellular bacteria via autophagy induction.

Deep Learning Network‐Tailored Microenvironment Matching of 4D Bioprinting Bioactive Scaffolds for Bone Regeneration

open access: yesAdvanced Science, EarlyView.
A DLN dataset was built to analyze MABS composition versus in vitro/in vivo osteogenesis and angiogenesis. An MLP neural network, taking BG morphological parameters as input, extracts bioactive features from these datasets. A rabbit tibial defect model then validates 4D‐printed MABS for adaptability and bone regeneration in critical defects.

SRAS1.1 E3 ligase mediates DSK2A degradation to regulate autophagy and drought tolerance in Arabidopsis

open access: yesEMBO Reports
Drought stress significantly impacts plant growth and productivity, requiring complex adaptive responses to ensure survival. In eukaryotes, autophagy and the ubiquitin-proteasome system (UPS) are critical pathways for maintaining cellular homeostasis ...

Porcine Deltacoronavirus M Protein Binds NLRP3 to Promote Inflammasome Assembly via Competition with TRIM31

open access: yesAdvanced Science, EarlyView.
Porcine deltacoronavirus (PDCoV) infection induces severe intestinal inflammation and acute diarrhea in piglets, yet the molecular mechanism remains incompletely understood. The M protein activates NLRP3 inflammasome through dual mechanisms: direct binding to the NLRP3 LRR domain and disruption of TRIM31‐mediated K48‐linked ubiquitination.

Penfluridol Triggers GSDME‐Mediated Immunogenic Pyroptosis to Potentiate Antitumor Immunotherapy

open access: yesAdvanced Science, EarlyView.
A high‐throughput screen of FDA‐approved antipsychotics identifies penfluridol as a potent pyroptosis inducer acting via direct TTI1 inhibition. This triggers TNFA‐NFKB signaling and caspase‐8/‐3‐dependent GSDME cleavage. The compound stimulates antitumor immunity alone and synergizes with anti‐PD‐1 therapy, while low TTI1 expression emerges as a ...

Introducing Borsantrazole: A Trifunctional Boron‐Based Pyrazole That Extends the Lifespan of Amyotrophic Lateral Sclerosis Mice

open access: yesAdvanced Science, EarlyView.
Herein we report a boron‐based pyrazole, (Borsantrazole ‐ a small molecule that selectively targets oxidative stress) that significantly increases survival, reduces weight loss, delays disease onset, and affects global protein changes in the SOD1‐G37R mouse model of ALS.

Inhalable ROS‐Responsive Nanospray Activates PPAR‐γ to Restore Macrophage Mitochondrial Homeostasis and Attenuate Radiation‐Induced Lung Injury

open access: yesAdvanced Science, EarlyView.
A nebulized nanoassembly alleviates radiation‐induced lung injury by reprogramming macrophages through coordinated ROS scavenging, NAD+ replenishment, and PPAR‐γ activation. Redox balance, mitochondrial homeostasis, and immune regulation are restored, revealing a microenvironment‐centered strategy for localized radioprotection.

Modulation of Lung Adenocarcinoma by Phosphorylated FOXN3‐Mediated Transcriptional Inactivation of p53

open access: yesAdvanced Science, EarlyView.
In non‐tumorous lung tissues, FOXN3 promotes the transcriptional activation of p53 by facilitating its recruitment to target promoters, thereby suppressing lung tumorigenesis through activation of the p53 signaling pathway. Conversely, in lung adenocarcinoma tissues, hyperphosphorylated FOXN3 dissociates from the promoters of p53‐responsive genes and ...
