Structural and biochemical basis for retinol‐binding protein 4 antagonism by tinlarebant

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 23, Page 6737-6748, December 2026.
Background and Purpose Retinol‐binding protein 4 (RBP4) is a member of the lipocalin superfamily that is connected to disease states such as insulin resistance, fatty liver disease and ocular disorders including Stargardt disease. Several retinoid and non‐retinoid antagonists of this protein have been developed for potential clinical use, but none have

Synthesis of Aminocyclopropanes by Redox‐Neutral Aminoalkylation

open access: yesAngewandte Chemie International Edition, Volume 65, Issue 45, 2 November 2026.
Aminocyclopropane synthesis by hydroaminoalkylation utilizing cheap, readily available starting materials is presented, offering orthogonality to commonly used methods and allowing broad functional‐group tolerance. Solvent‐mediated conditions enable the reaction of a pre‐formed hemiaminal with alkenes and alkynes.

Neurotransmitter‐Defined Degeneration Patterns in Sporadic and C9orf72‐Associated Amyotrophic Lateral Sclerosis: Predilection to GABAergic, Serotonergic, Opioid, Glutamatergic, Endocannabinoid, and Microglial Systems—Implications for Therapy Development

open access: yesAnnals of Neurology, Volume 100, Issue 5, Page 948-961, November 2026.
Objective Amyotrophic lateral sclerosis (ALS) has a markedly distinctive clinical and neuroradiological signature, with the preferential involvement of specific brain networks and the apparent sparing of others. The molecular underpinnings of the strikingly selective anatomical vulnerability have not been fully elucidated to date despite the potential ...

Improved Serotonin Measurement With Fast‐Scan Cyclic Voltammetry: Analysis of Electrode Fouling by Fluoxetine in Live Brain Tissue

open access: yesAnalysis &Sensing, Volume 6, Issue 6, November 2026.
Fluoxetine (Prozac) is a drug that is useful for treating depression and for studying serotonin system function in live brain tissue. Unfortunately, the use of fluoxetine tends to foul electrodes when using fast‐scan cyclic voltammetry. In this work, we evaluated an extended serotonin waveform as a useful approach for mitigating fouling by fluoxetine ...

Do Brain Extracellular Fluid Concentrations Predict PET‐Inferred Receptor Occupancy in Human CNS? A Mechanistic Model‐Based Analysis of Dopamine D2 Receptor Ligands

open access: yesClinical Pharmacology &Therapeutics, Volume 120, Issue 5, Page 1389-1401, November 2026.
A mechanistic CNS PBPK model framework was applied to eight antipsychotic D2 receptor ligands, comparing brain extracellular fluid (brainECF) and brain cell membrane (brainM) as biophase for predicting human receptor occupancy compared against PET neuroimaging data. BrainM concentrations showed improved agreement with PET‐derived receptor occupancy for
