Severe asthma: oral corticosteroid alternatives and the need for optimal referral pathways

open access: yesJournal of Asthma, 2020
Objective Patients with severe asthma require high-dose inhaled corticosteroids, with or without add-on treatments, to maintain asthma control. Because symptom control remains unsatisfactory in some patients despite these therapies, maintenance therapy ...

Inhibition of RACK1‐Mediated NLRP3 Oligomerization (Active Conformation) Ameliorates Acute Respiratory Distress Syndrome

open access: yesAdvanced Science, EarlyView.
Schematic diagram showing the potential mechanism of bigelovin on the activation of NLRP3 inflammasome Bigelovin may inhibit activated protein C kinase 1 (RACK1) by directly binding with cys168 of RACK1. Bigelovin thus prevents oligomerization of NLRP3 (NLRP3 active conformation) and subsequent assembly of NLRP3 inflammasome, blocking the activation of

Severe Asthma in Adults

open access: yesAmerican Journal of Respiratory and Critical Care Medicine, 2005
Severe asthma remains poorly understood and frustrating to care for, partly because it is a heterogeneous disease. Patients with severe asthma disproportionately consume health care resources related to asthma. Severe asthma may develop over time, or shortly after onset of the disease.

Azithromycin and severe asthma [PDF]

open access: yesThorax, 2013
Brusselle et al 1 performed an interesting academic-based randomised controlled trial Azithromycin for Prevention of Exacerbations in Severe Asthma (AZISAST) of azithromycin 250 mgm taken thrice weekly versus placebo as add-on therapy for 26 weeks in non-smoking patients with severe asthma, defined as Global Initiative for Asthma (GINA) steps 4–5 ...

Eosinophils‐Induced Lumican Secretion by Synovial Fibroblasts Alleviates Cartilage Degradation via the TGF‐β Pathway Mediated by Anxa1 Binding

open access: yesAdvanced Science, EarlyView.
Eosinophils play a crucial role in the progression of temporomandibular joint osteoarthritis (TMJOA). This study demonstrates that eosinophils, induced by OVA/IL‐5, promote Lumican secretion in the synovium, which binds to Annexin A1 in chondrocytes, inhibiting transforming growth factor β2 and Smad2/3 phosphorylation.

Severe asthma in Japan

open access: yesAllergology International, 2019
The characteristic phenotype of severe asthma in Japan seems to be distilled into the following two features: low incidence of obesity and high prevalence of patients with type 2 inflammation. Only 5-7% of Japanese severe asthma patients had a body mass index (BMI) ≥30 kg/m2, and more than 80% of patients with severe asthma exhibited type 2 ...

Dupilumab efficacy in uncontrolled, moderate-to-severe asthma with self-reported chronic rhinosinusitis [PDF]

open access: yes, 2019
BACKGROUND: Dupilumab, a fully human monoclonal antibody, blocks the shared receptor component for IL-4 and IL-13 signaling, key drivers of type 2 inflammation.

Biologics and global burden of asthma: A worldwide portrait and a call for action

open access: yesWorld Allergy Organization Journal, 2021
Biologics for severe asthma can significantly impact on the burden of disease and also have the potential to reduce asthma mortality. By reviewing the literature and contacting the pharmaceutical companies, the present paper aims at providing a worldwide

Environmental allergen reduction in asthma management: an overview

open access: yesFrontiers in Allergy, 2023
Asthma is a prevalent non-communicable disease that affects both children and adults. Many patients with severe, uncontrolled asthma could not achieve total control despite using anti-asthmatic drugs.
