Glioma cells mainly express the endothelin receptor EDNRB, while EDNRA is restricted to a perivascular tumor subpopulation. Endothelin signaling reduces glioma cell proliferation while promoting migration and a proneural‐to‐mesenchymal transition associated with poor prognosis. This pathway activates Ca2+, K+, ERK, and STAT3 signalings and is regulated

Directed evolution and structural analysis of an OB-fold domain towards a specifc binding reagent
Interactions between proteins are a central concept in biology, and understanding and manipulation of these interactions is key to advancing biological science.

While covalent drug discovery is reemerging as an important route to small molecule therapeutic leads, strategies for the discovery and engineering of protein-based irreversible binding agents remain limited. Here, we describe the use of yeast display, a

Monoclonal Antibodies that Inhibit the Proteolytic Activity of Botulinum Neurotoxin Serotype/B
Existing antibodies (Abs) used to treat botulism cannot enter the cytosol of neurons and bind to botulinum neurotoxin (BoNT) at its site of action, and thus cannot reverse paralysis.

Hippo pathway at the crossroads of stemness and therapeutic resistance in breast cancer
Dysregulation of the Hippo pathway drives nuclear accumulation of YAP/TAZ, activating stemness‐related transcriptional programs that sustain breast cancer stemness and fuel therapeutic resistance across subtypes, underscoring Hippo signaling as a targetable vulnerability. Figure created and edited with BioRender.com.

CCDC80 suppresses high‐grade serous ovarian cancer migration via negative regulation of B7‐H3
PAX8 is a lineage‐specific master regulator of transcription in high‐grade serous ovarian cancer (HGSC) progression. We show for the first time that PAX8 facilitates proliferation and metastasis by repressing the cell autonomous tumor suppressor CCDC80 and inducing B7‐H3 expression.

Structural and biochemical analysis of the UvrA binding module of the bacterial transcription repair coupling factor Mfd [PDF]
The Mfd (mutation frequency decline) protein is responsible for connecting the cellular processes of transcription and DNA repair in bacteria. Mfd, also termed transcription-repair coupling factor (TRCF), recognizes arrested transcription elongation ...

Single-chain antibody sequences and structural model.
Three novel phage-displayed single-chain antibodies were identified via biopanning and named NJT-R3-A1, NJT-R3-A2, and NJT-R3-A3 based on their abilities to bind the NV P-domain after three rounds of binding selection (A).

Structural studies on protein scaffolds related to muscle physiology and disease : the titin filament, its associated component MuRF-1 and nuclear LAP2[alpha] [PDF]
The titin molecule has a length of overµm and functions as a colossal protein scaffold in the muscle sarcomere. Up to 90% of its total mass is composed of repetitive immunoglobulin (Ig) and fibronectin (FnIII) domains that form linear tandems ...
