Identification of Critical Antibody-Binding Sites in the HIV-1 gp41 Six-Helix Bundle Core as Potential Targets for HIV-1 Fusion Inhibitors

open access: yesImmunobiology, 2009
Formation of the six-helix bundle (6-HB) core between the N- and C-terminal heptad repeats (NHR and CHR) regions of the HIV-1 envelope glycoprotein (Env) transmembrane subunit gp41 is a critical step during the process of virus and target cell membrane fusion.

The Fusion Core Complex of the Peste des Petits Ruminants Virus Is a Six-Helix Bundle Assembly

Biochemistry, 2003
We describe the properties of the two heptad repeats (HR1 and HR2) of the Peste des petits ruminants virus (PPRV) fusion protein (F) to obtain insights into the mechanism by which these repeats influence PPRV-mediated cell fusion. Both HR1 and HR2 inhibit PPRV-mediated syncytia formation in Vero cells in vitro.

AI-Guided Hydrophobic Core Design of Robust Six-Helix Bundle Proteins

ACS Nano
α-Helical domains are widespread and versatile, yet typically fail under low mechanical load because backbone hydrogen bonds unzip sequentially, limiting their use in force-bearing nanomaterials and molecular devices. We present an AI-guided strategy to design six-helix bundle proteins with densely packed hydrophobic cores that co-optimize mechanical ...

Broad-spectrum coronavirus inhibition by RSV fusion inhibitors targeting six-helix bundle formation

Life Sciences
Waning vaccine-derived immunity and the emergence of new SARS-CoV-2 subvariants continue to challenge global public health, emphasizing the need for novel inhibitors against SARS-CoV-2 infection. Molecular inhibitors targeting virus entry are promising candidates as they can disrupt viral replication at the earliest stage.

Impact of the Enfuvirtide Resistance Mutation N43D and the Associated Baseline Polymorphism E137K on Peptide Sensitivity and Six-Helix Bundle Structure

Biochemistry, 2008
Enfuvirtide (ENF), the first human immunodeficiency virus type 1 (HIV-1) fusion inhibitor approved for clinical use, acts by binding to gp41 heptad repeat 1 (HR1) and preventing its interaction with the viral HR2 region. Treatment-emergent resistance to ENF has been mapped to residues within HR1, and these mutations decrease its susceptibility to ENF ...

Evolutionary Repair of HIV Type 1 gp41 with a Kink in the N-Terminal Helix Leads to Restoration of the Six-Helix Bundle Structure

AIDS Research and Human Retroviruses, 2004
The HIV-1 envelope glycoprotein complex (Env) can be stabilized by the introduction of a disulfide bond between the gp120 and gp41 subunits. The resulting protein is monomeric, but trimerization can be improved by the introduction of a single helix-breaking residue at the conserved Ile559 site in the N-terminal heptad repeat region of gp41.

The Hydrophobic Pocket Contributes to the Structural Stability of the N-Terminal Coiled Coil of HIV gp41 but Is Not Required for Six-Helix Bundle Formation

Biochemistry, 2003
In models of HIV fusion, the glycoprotein gp41 is thought to form a six-helix bundle during viral fusion with the target cell. This bundle is comprised of three helical regions (from the heptad repeat 2, or HR2, region of gp41) bound to an inner, trimeric, coiled-coil core (from the HR1 region).

Identification of inhibitors of the HIV-1 gp41 six-helix bundle formation from extracts of Chinese medicinal herbs Prunella vulgaris and Rhizoma cibotte

Life Sciences, 2002
An increasing portion of patients with HIV infection and/or AIDS cannot use currently FDA-approved anti-HIV drugs, including the reverse transcriptase and protease inhibitors, due to the adverse effects and the emergence of drug resistance. Thus, it is essential to develop new anti-HIV agents with a target different from the HIV reverse transcriptase ...

Prospects and strategies for the discovery and development of small-molecule inhibitors of six-helix bundle formation in class 1 viral fusion proteins.

Current opinion in investigational drugs (London, England : 2000), 2006
Class I viral fusion proteins have an important role in the fusion of viral membranes with host cell membranes, a critical step in the viral life-cycle. These proteins all have similar structural features and form six-helix bundles in their fusogenic form, a general mechanism of action for virus-cell fusion.
