Physical frailty predicts adverse outcomes in hospitalized patients with acute decompensation of cirrhosis

open access: yes
reservedBACKGROUND AND AIM: Patients with end-stage liver disease are at high risk of malnutrition, sarcopenia and frailty. Recent evidence indicates that physical frailty, as assessed by liver frailty index (LFI), is an independent predictor of ...

Transient Sleep Deprivation Induces Persistent Auditory Neuropathy via ROS‐Initiated Neuroinflammation and BK Channel Suppression

open access: yesAdvanced Science, EarlyView.
Sleep disturbance severity closely tracks hearing loss in a clinical cohort, yet the mechanistic link remains unclear. Acute sleep deprivation is shown to trigger transient cochlear oxidative stress that switches into a self‐sustaining neuroinflammatory state, suppressing BK channels and causing irreversible synaptopathy.

Influence of ACE inhibitors on frailty, cardiac contractile function and calcium homeostasis in middle-aged male and female mice

open access: yes, 2016
Angiotensin Converting Enzyme (ACE) inhibitors were chronically administered to middle-aged male and female mice to determine whether treatment with ACE inhibitors attenuates frailty and whether this is accompanied by changes in cardiac function.

A Dual‐Membrane Biomimetic Nanoplatform Enables Triple‐Modal Therapy Against SARS‐CoV‐2 Through Viral Decoy, Inflammation Neutralizing, and Intracellular RNAi

open access: yesAdvanced Science, EarlyView.
[A&T]MLN is a triple‐modal nanoplatform with siRNA‐loaded LN coated with hybrid ACE2/macrophage membrane. It blocks viral entry, neutralizes IL‐6/IL‐1β/TNF‐α, and delivers siRNA to suppress viral replication. In a murine lung injury model, it attenuates inflammation, offering a multi‐pronged strategy against SARS‐CoV‐2 variants and hyperinflammation ...

TGFBI Maintains Fibrocartilage Homeostasis by Restraining O‐GlcNAcylation to Inactivate and Destabilize ADAMTS16 in Temporomandibular Joint Osteoarthritis

open access: yesAdvanced Science, EarlyView.
TGFBI deficiency elevates O‐GlcNAcylation, promoting the transcriptional activation of ADAMTS16 and OGT‐mediated post‐translational modification of ADAMTS16 at Ser1170. This leads to its stabilization and accumulation, driving chondrocyte metabolic disorders and senescent phenotypes, ultimately resulting in fibrocartilage dysfunction and TMJOA ...

Upadacitinib Restrains the Pathogenic Fitness of CD4+ T Cells and Aberrant B Cell Programming in Optic Neuritis

open access: yesAdvanced Science, EarlyView.
Single‐cell profiling and functional perturbation reveal coordinated JAK1‐pSTAT3 downstream programs in optic neuritis, including MCL1‐dependent fitness of pathogenic CD4+ Tem cells and glycolysis‐linked, cholesterol‐sensitive B‐cell responses associated with RORA. Upadacitinib disrupts this reciprocal T‐B‐cell circuit and alleviates neuroinflammation,
