Mutagenic effect of borocaptate sodium and boronophenylalanine in neutron capture therapy

International Journal of Radiation Oncology Biology Physics, 2002
To investigate the mutagenic effect in boron neutron capture therapy (BNCT), Chinese hamster ovary cells were incubated with 10 B-enriched borocaptate sodium (BSH) or para-boronophenylalanine (BPA) before exposure to thermal neutrons, and the occurrence of mutations at the hypoxanthine-guanine phosphoribosyl transferase (HPRT) locus was measured.BSH or

11B nuclear magnetic resonance studies of the interaction of borocaptate sodium with serum albumin

Biochemical Pharmacology, 1995
The interaction between borocaptate sodium, Na2B12H11SH (BSH), and three types of serum albumin--bovine, human and dog (BSA, HSA and DSA)--has been investigated quantitatively using 11B NMR. The 11B chemical shifts and relaxation rates of BSH were studied with various concentrations of serum albumin (1-5%, w/v) at 295-310 degrees K.

Pharmacokinetics of sodium borocaptate: a critical assessment of dosing paradigms for boron neutron capture therapy

Journal of Neuro-Oncology, 2003
The pharmacokinetics of sodium borocaptate (BSH), a drug that has been used clinically for boron neutron capture therapy (BNCT) of malignant brain tumors, have been characterized by measuring boron concentrations by direct current plasma-atomic emission spectroscopy (DCP-AES) in a group of 23 patients with high-grade gliomas.

Boron neutron capture irradiation of the rat spinal cord: effects of variable doses of borocaptate sodium

Radiotherapy and Oncology, 1996
The Fischer 344 rat spinal cord model has been used to evaluate the response of the central nervous system to boron neutron capture irradiation with variable doses of the neutron capture agent, borocaptate sodium (BSH). Three doses of BSH, 190, 140 and 80 mg/kg body weight, administered by i.p.

Enhancement of sodium borocaptate (BSH) uptake by tumor cells induced by glutathione depletion and its radiobiological effect

Cancer Letters, 2004
Sodium borocaptate (BSH) is widely used for boron neutron capture therapy (BNCT) of brain tumors. However, the mechanism of uptake by the tumor remains unclear. We investigated the sulfhydryl moiety of this compound. Down regulation of glutathione (GSH) by buthionine sulfoximine in cultured cells resulted in increase of BSH uptake (7.9-36.5%) compared ...

Borocaptate Sodium (BSH) Pharmacokinetics in Glioma Patients

1992
In a previous communication we have reported the results of a biodistribution study of intravenous borocaptate sodium (BSH) administered to 28 patients undergoing craniotomy for various intracranial tumors10. The measured boron concentrations in tumor, blood, and normal cranial tissues led to the tentative conclusion 1) that BSH readily enters most ...

Response of rat skin to boron neutron capture therapy with p-boronophenylalanine or borocaptate sodium

Radiotherapy and Oncology, 1994
The effects of boron neutron capture irradiation employing either BPA or BSH as neutron capture agents has been assessed using the dorsal skin of Fischer 344 rats. Pharmacokinetic studies, using prompt gamma spectrometry, revealed comparable levels of boron-10 (10B) in blood and skin after the intravenous infusion of BSH (100 mg/kg body wt.).

The usefulness of 2-nitroimidazole-sodium borocaptate-10B conjugates as 10B-carriers in boron neutron capture therapy

Applied Radiation and Isotopes, 2004
We evaluated the usefulness of five new (10)B-compounds (TX-2016, TX-2017, TX-2018, TX-2041, and TX-2042) as (10)B-carriers in boron neutron capture therapy (BNCT). They are 2-nitroimidazole-sodium borocaptate-(10)B (BSH) conjugates, that is, hybrid compounds that have both hypoxic tumor cell sensitizing unit under gamma-ray irradiation, 2 ...

Disposition and tissue distribution of boron after infusion of borocaptate sodium in patients with malignant brain tumors

International Journal of Radiation Oncology*Biology*Physics, 1998
In the frame of the Czech boron neutron capture therapy (BNCT) project, a clinical Phase I study of borocaptate sodium [Na2B12H11SH (BSH)] as the boron-10 delivery agent was performed to obtain data on disposition and tissue distribution of boron after an infusion of this compound, as well as to establish an optimal protocol for BNCT of malignant ...
