The purification scheme utilizes ultrafiltration through membranes of nominal mol. wt. cutoffs, molecular sieve chromatography and finally iso-electric focusing.

The sensitivity of the assay permits the measurement of this factor in human plasma diluted 1:20,000 and in monkey plasma diluted 1:5000. Somatomedin B is not detected in many nonprimate plasmas diluted 1:20.

Circulating somatomedin activity reflects the presence of both somatomedins and somatomedin inhibitors, factors which antagonize the growth-promoting actions of somatomedins. Although both are regulated by nutrition, somatomedin inhibitors respond more rapidly than somatomedins to refeeding in fasted animals.

The biosynthesis of somatomedins and their role in the fetus
