Targeting sphingosine kinase 1 in cancer

Advances in Biological Regulation, 2012
Cancer is one of largest killers in the world and defeating it represents one of the most important challenges in Medicine today. For instance, annually, around 1.38 million women are diagnosed worldwide with breast cancer and approximately 0.25 million men worldwide are diagnosed with prostate and testicular cancer.

Sphingosine kinase 1 in viral infections

Reviews in Medical Virology, 2012
SUMMARYSphingosine kinase 1 (SphK1) is an enzyme that phosphorylates the lipid sphingosine to generate sphingosine‐1‐phosphate (S1P). S1P can act intracellularly as a signaling molecule and extracellularly as a receptor ligand. The SphK1/S1P axis has well‐described roles in cell signaling, the cell death/survival decision, the production of a pro ...

Post-translational regulation of sphingosine kinases

Biochimica et Biophysica Acta (BBA) - Molecular and Cell Biology of Lipids, 2013
Sphingosine kinases (SKs) catalyse the conversion of sphingosine to sphingosine 1-phosphate (S1P), a signalling lipid that is involved in a plethora of cellular processes including proliferation, apoptosis, calcium homeostasis, angiogenesis, vascular and neuronal maturation, cell migration and immune responses.

Regulation of sphingosine kinase and sphingolipid signaling

Trends in Biochemical Sciences, 2011
Bioactive sphingolipids, including ceramide, sphingosine and sphingosine 1-phosphate are important regulators of many cellular processes, including cell survival, proliferation, differentiation, migration and immune responses. Although the levels of these bioactive sphingolipids are regulated by complex pathways subject to spatial and temporal control,

Sphingosine kinase inhibitors

2019
Sphingosine kinases are enzymes that catalyze the biosynthesis of sphingosine-1-phosphate. The invention provides compounds that are effective for inhibition of sphingosine kinase type 1, sphingosine kinase type 2, or both. Certain compounds are selective for sphingosine kinase type 2 relative to sphingosine kinase type 1.
