ABSTRACT Background Aberrant T‐helper 1 (Th1) and Th17 cell activation is involved in the pathogenesis of inflammatory bowel disease (IBD). Psychosine is known to induce apoptosis, promote astrocyte activation, and inhibit osteoclastogenesis. However, the role of psychosine on Th1 and Th17 cell immune responses has not been previously reported. In this

Pancreatic cancer is known to be the most lethal cancer. Fewer new treatments are being developed for pancreatic cancer as compared to other cancers. The bioactive lipid S1P, which is mainly regulated by sphingosine kinase 1 (SK1) and sphingosine kinase ...

A concomitant action of multiple profibrotic mediators appears crucial in the development and progression of fibrosis. Sphingosine kinase/sphingosine 1 phosphate and transforming growth factor-β/Smads pathways are both involved in pathogenesis of ...

A Somatic Mutation in HIF‐2α Affects Glucose and Lipid Metabolism via CD36 and Ceramide
A mutation within the C‐terminal transactivation domain of HIF‐2α enhances Cd36 expression by increased transactivation activity, leading to ceramide accumulation and disruption of glucose and lipid metabolism. Inhibition of ceramide synthesis by myriocin reduces ceramide levels and ameliorates these metabolic disturbances.

The infiltration of melanoma tumors by macrophages is often correlated with poor prognosis. However, the molecular signals that regulate the dialogue between malignant cells and the inflammatory microenvironment remain poorly understood.

Flow dynamics control the effect of sphingosine-1-phosphate on endothelial permeability in a microfluidic vessel bifurcation model [PDF]
Blood vessels are lined by endothelial cells that form a semipermeable barrier to restrict fluid flow across the vessel wall. The endothelial barrier is known to respond to various molecular mechanisms, but the effects of mechanical signals that arise ...

The role of sphingosine 1-phosphate (S1P) in liver fibrosis or inflammation was not fully examined in human. Controversy exists which S1P receptors, S1P1 and S1P3 vs S1P2, would be importantly involved in its mechanism.

Role of sphingosine kinase and sphingosine-1-phosphate in inflammatory arthritis
The importance of sphingosine kinase (SphK) and sphingosine-1-phosphate (S1P) in inflammation has been extensively demonstrated. As an intracellular second messenger, S1P plays an important role in calcium signaling and mobilization, and cell proliferation and survival.
