Psilocybin as Transformative Fast‐Acting Antidepressant: Pharmacological Properties and Molecular Mechanisms

open access: yesFundamental &Clinical Pharmacology, Volume 39, Issue 4, August 2025.
ABSTRACT In the 1950s–60s, serotonergic psychedelic drugs were studied as potential adjuvants to psychotherapy to treat addiction and alcoholism. However, starting in the 70s, preclinical and clinical studies on psychedelics stopped for decades because legislation controlled its recreational use, citing their hallucinogenic and psychotomimetic effects,

Zebrafish Behavioral Profiling Links Drugs to Biological Targets and Rest/Wake Regulation [PDF]

open access: yes, 2010
A major obstacle for the discovery of psychoactive drugs is the inability to predict how small molecules will alter complex behaviors. We report the development and application of a high-throughput, quantitative screen for drugs that alter the behavior ...

Synthesis of Pyrrolidin‐3‐Ones and Spiropyrrolidinones by 1,3‐Dipolar Cycloadditions of Nitrones with Allenylphosphonates

open access: yesEuropean Journal of Organic Chemistry, Volume 28, Issue 27, July 25, 2025.
This work introduces an efficient and gram‐scale microwave‐assisted (3+2) cycloaddition of nitrones with fluorinated allenylphosphonates, yielding diverse 4‐phosphinyl‐pyrrolidin‐3‐ones with exclusive 4,5‐trans diastereoselectivity in moderate to good yields.

Pharmacological profile of novel psychoactive benzofurans [PDF]

open access: yes, 2015
Benzofurans are newly used psychoactive substances, but their pharmacology is unknown. The aim of the present study was to pharmacologically characterize benzofurans in vitro.; We assessed the effects of the benzofurans 5-APB, 5-APDB, 6-APB, 6-APDB, 4 ...

Current status and features of antipsychotic prescriptions in Japanese forensic psychiatric wards based on a forensic inpatient database

open access: yesNeuropsychopharmacology Reports, Volume 45, Issue 1, March 2025.
We analyzed the current status of antipsychotics for Japanese forensic inpatients with schizophrenia spectrum disorders nationwide. Olanzapine was the most commonly prescribed antipsychotic, followed by clozapine. Clozapine was associated with longer hospitalization and a higher rate of seclusion experience.

Monoamine receptor interaction profiles of 4-thio-substituted phenethylamines (2C-T drugs) [PDF]

open access: yes, 2017
4-Thio-substituted phenethylamines (2C-T drugs) are potent psychedelics with poorly defined pharmacological properties. Because of their psychedelic effects, 2C-T drugs are sometimes sold as new psychoactive substances (NPSs).

Aromatic linker variations in novel dopamine D2 and D3 receptor ligands

open access: yesArchiv der Pharmazie, Volume 357, Issue 8, August 2024.
Within the blueprint motifs of dopamine D2R/D3R ligands, the butyl (tetramethylene) linker was replaced by substituted benzyl or phenethyl linkers, maintaining high affinities at both receptor subtypes. Abstract Dopamine D2‐like receptors, especially D2 and D3 receptor subtypes, are important targets of antipsychotic agents.

Discovery of a Small Molecule Activator of Slack (Kcnt1) Potassium Channels That Significantly Reduces Scratching in Mouse Models of Histamine‐Independent and Chronic Itch

open access: yesAdvanced Science, Volume 11, Issue 15, April 17, 2024.
The potassium channel Slack is highly expressed in sensory neurons that sense itch. In this study, several new compounds that activate Slack are developed. It is found that the lead molecule, compound 6, effectively inhibits scratching elicited by multiple pruritogens in mice establishing Slack activation as a novel pharmacological strategy for the ...

Blockade D2 dopamine receptors as a new approach to correction of violations in alveolar parenchyma and regulation of endothelial progenitor cells in pulmonary emphysema [PDF]

open access: yes, 2018
On the model of emphysema of the lungs caused by an extract of cigarette smoke or elastase, it was shown that spiperone has an anti-inflammatory effect, reduces the area of emphysema, stimulates the regeneration of alveolar cells and the microvascular ...
