Unconventional Lysine-Type Lipid Assemblies Enable Efficient Antisense Oligonucleotide Delivery with Distinct Structural Features

open access: yesPharmaceutics
Background/Objectives: Antisense oligonucleotides (ASOs) hold great therapeutic potential due to their precise ability to modulate gene expression, particularly for treating genetic and neurological disorders.

Modulating the influenza A virus – target membrane fusion interface with synthetic DNA-lipid receptors [PDF]

open access: yes, 2021
Influenza A virus (IAV) binds to sialylated glycans on the cell membrane before endocytosis and fusion. Cell surface glycans are highly heterogenous in length and glycosylation density, which leads to variation in the distance and rigidity with which IAV

Processing DNA Storage through Programmable Assembly in a Droplet‐Based Fluidics System

open access: yesAdvanced Science, 2023
DNA can be used to store digital data, and synthetic short‐sequence DNA pools are developed to store high quantities of digital data. However, synthetic DNA data cannot be actively processed in DNA pools.

Shuffling of DNA Cassettes in a Synthetic Integron [PDF]

open access: yes, 2013
The complexity of even small gene networks makes them hardly amenable to rational design. Testing random combinations of genetic elements in a directed evolution procedure is thus of interest for many applications including metabolic engineering. Here we describe how the recombination machinery of class 1 integrons can be used to deliver and shuffle ...

Autonomous assembly of synthetic oligonucleotides built from an expanded DNA alphabet. Total synthesis of a gene encoding kanamycin resistance

open access: yesBeilstein Journal of Organic Chemistry, 2014
Background: Many synthetic biologists seek to increase the degree of autonomy in the assembly of long DNA (L-DNA) constructs from short synthetic DNA fragments, which are today quite inexpensive because of automated solid-phase synthesis.

DNA Photonics – Photoinduced Electron Transfer in Synthetic DNA-Donor–Acceptor Systems

open access: yesCHIMIA, 2007
Recent experimental and theoretical results have demonstrated that structural dynamics are critical for a comprehensive mechanistic understanding of DNA charge transfer (CT).

Gene transfer into hepatocytes using asialoglycoprotein receptor mediated endocytosis of DNA complexed with an artificial tetra-antennary galactose ligand [PDF]

open access: yes, 1992
We have constructed an artificial ligand for the hepatocyte-specific asialoglycoprotein receptor for the purpose of generating a synthetic delivery system for DNA.

Gut microbiome and aging—A dynamic interplay of microbes, metabolites, and the immune system

open access: yesFEBS Letters, EarlyView.
Age‐dependent shifts in microbial communities engender shifts in microbial metabolite profiles. These in turn drive shifts in barrier surface permeability of the gut and brain and induce immune activation. When paired with preexisting age‐related chronic inflammation this increases the risk of neuroinflammation and neurodegenerative diseases.

Synthetic DNA for Cell‐Surface Engineering

open access: yesAngewandte Chemie, 2021
AbstractThe cell membrane is not only a physical barrier, but also a functional organelle that regulates the communication between a cell and its environment. The ability to functionalize the cell membrane with synthetic molecules or nanostructures would advance cellular functions beyond what evolution has provided.

Hyperosmotic stress induces PARP1‐mediated HPF1‐dependent mono(ADP‐ribosyl)ation

open access: yesFEBS Letters, EarlyView.
Sorbitol‐induced hyperosmotic stress rapidly induces reversible mono(ADP‐ribosyl)ation (MARylation) on PARP1 without the signs of genotoxic signaling. We show that PARP1 autoMARylation is HPF1 dependent and forms hydroxylamine‐resistant O‐glycosidic linkages.
