Catalytic Control in the Facile Proton Transfer in Taxadiene Synthase

ACS Catalysis, 2017
Enzymes are highly efficient and usually very specific biocatalysts. However, some enzyme families, such as terpene synthases, are inherently promiscuous due to the extremely challenging chemistry they have evolved to tackle. Here we focus on one such enzyme, taxadiene synthase (TXS), which produces taxa-4(5),11(12)-diene, a key precursor to the ...

Taxadiene Synthase-Catalyzed Cyclization of 6-Fluorogeranylgeranyl Diphosphate to 7-Fluoroverticillenes

Journal of the American Chemical Society, 2005
The mechanism of the taxadiene synthase-catalyzed cyclization of (E,E,E)-geranylgeranyl diphosphate (GGPP, 7) to taxadiene (5) is proposed to proceed through a verticillen-12-yl carbocation intermediate (8) that undergoes an 11 --> 7 proton transfer leading to formation of the C ring.

[Metabolomics analysis of taxadiene producing yeasts].

Sheng wu gong cheng xue bao = Chinese journal of biotechnology, 2015
In order to study the inherent difference among terpenes producing yeasts from the point of metabolomics, we selected taxadiene producing yeasts as the model system. The changes of cellular metabolites during fermentation log phase of artificial functional yeasts were determined using metabolomics methods.

Unraveling the multispecificity and catalytic promiscuity of taxadiene monooxygenase

Journal of Molecular Catalysis B: Enzymatic, 2014
The enzyme taxadiene synthase of the paclitaxel biosynthetic pathway cyclizes geranylgeranyl pyrophosphate (GGPP) to produce taxadiene, which is then believed to be stereo- and regiospecifically oxidized by taxadiene 5α-hydroxylase to form taxadien-5α-ol. However, when the metabolism of Escherichia coli was re-engineered in the present study to express

Mechanistic Studies of Taxadiene and Abietadiene Synthases

2001
Abstract : Investigation aimed at understanding the enzymatic mechanism of the two diterpene cyclases, taxadiene and abietadiene synthase, may yield an alternative approach to combinatorial chemistry in the search for new and improved chemotherapeutic and antibiotic drugs.
