After activation, CD4 helper T (Th) cells differentiate into Th1or Th2 effector subsets. These two types of cells produce distinct profiles of cytokines and regulate different immune responses. This review summarizes recent progress on different features of co-stimulatory regulation and chemokine-mediated homing of Th1 and Th2 cells.

Although immunologists have found the dichotomous helper T cell subset model a useful construct, it is beginning to show its age. Some suggest that proponents of the TH1-TH2 dichotomy are overzealous in making the data conform to the scheme, like Procrustes who stretched or truncated his guests so that they would fit his guest bed.

Th1 lymphocytes secrete interferon gamma (IFNγ) and lymphotoxin, promoting cell-mediated immunity to intracellular pathogens; and Th2 lymphocytes secrete interleukins 4 and 5 (IL-4 and IL-5), which function in allergy and humoral immunity to parasites.
