Effects of the neutral endopeptidase inhibitor thiorphan on cardiovascular and renal function in cirrhotic rats [PDF]

open access: bronzeBritish Journal of Pharmacology, 2003
Cirrhosis is associated with cardiovascular and renal dysfunction including sodium retention. Many vasoactive peptides such as atrial natriuretic peptide (ANP) and endothelin‐1 (ET‐1) are degraded by neutral endopeptidase 24.11 (NEP). We investigated the hemodynamic and renal effects of thiorphan, a NEP inhibitor, in a rat cirrhosis model.

The role of bestatin-sensitive aminopeptidase, angiotensin converting enzyme and thiorphan-sensitive "enkephalinase" in the potency of enkephalins in the guinea-pig ileum.

open access: diamondJapanese Journal of Pharmacology, 1984
The role of each enkephalin-hydrolyzing peptidase in the inhibitory potency of exogenously added enkephalins in the myenteric plexus-longitudinal muscle preparation of guinea-pig ileum was studied by using the relatively specific inhibitor of each enzyme.

The inactivation of [Met5]-enkephalin by bestatin-sensitive aminopeptidase, captopril-sensitive peptidyl dipeptidase A and thiorphan-sensitive endopeptidase-24.11 in mouse vas deferens.

open access: diamondJapanese Journal of Pharmacology, 1986
The enkephalin-hydrolyzing peptidases in mouse vas deferens were studied and compared with those in guinea pig ileum which had been characterized in the previous study.

Relationship between enkephalinase inhibition of thiorphan in vivo and its analgesic activity.

open access: bronzeJournal of Pharmacobio-Dynamics, 1985
The relationship between enkephalinase inhibition by thiorphan in vivo and analgesic activity in nociceptive tests was studied. The analgesic activity of thiorphan in various nociceptive tests was compared with that of a narcotic analgesic, morphine and antipyretic analgesic, antipyrine.

Ligand Screening and Discovery Using Cocktail Soaking and Automated Microcrystal Electron Diffraction. [PDF]

open access: yesChemMedChem
Cocktail soaking using single‐crystal X‐ray diffraction allows high‐throughput crystallographic ligand screening against protein targets. However, protein microcrystals are not amenable to this approach. Herein, a workflow integrating cocktail soaking with automated microcrystal electron diffraction to allow rapid ligand screening, structure ...

Design, synthesis, and evaluation of novel racecadotril-tetrazole-amino acid derivatives as new potent analgesic agents

open access: yesResearch in Pharmaceutical Sciences, 2021
Background and purpose: Although pain is one of the most common symptoms of diseases, it is often mismanaged due to limited access to painkillers and ineffectiveness, unacceptable side effects, or the possibility of abuse.

Granulocyte-colony stimulating factor attenuates oligomeric amyloid β neurotoxicity by activation of neprilysin. [PDF]

open access: yesPLoS ONE, 2014
Soluble oligomeric amyloid β (oAβ) causes synaptic dysfunction and neuronal cell death, which are involved in the pathogenesis of Alzheimer's disease (AD).

Concomitant medication in patients with bradykinin‐mediated angioedema – there's more than ACE inhibitors

open access: yesJDDG: Journal der Deutschen Dermatologischen Gesellschaft, Volume 21, Issue 11, Page 1283-1289, November 2023., 2023
Summary Bradykinin‐mediated angioedema is a rare, non‐allergic, potentially life‐threatening disease. ACE inhibitor‐induced angioedema and hereditary angioedema (HAE) are the two most common presentations. Therapeutic options, pathophysiology and diagnosis continue to be investigated, with considerable progress in HAE over the last few decades. For all
