Targeting Tn-Antigen-Positive Human Tumors with a Recombinant Human Macrophage Galactose CType Lectin

open access: yes, 2021
Alterations in glycosylation cause the emergence of tumor-associated carbohydrate antigens (TACAs) during tumorigenesis. Truncation of O-glycans reveals the Thomsen nouveau (Tn) antigen, an N-acetylgalactosamine (GalNAc) frequently attached to serine or ...

Side population cells derived from hUCMSCs and hPMSCs could inhibit the malignant behaviors of Tn+ colorectal cancer cells from modifying their O-glycosylation status

open access: yesStem Cell Research & Therapy, 2023
Background Cosmc (C1GalT1C1) mutation could cause aberrant O-glycosylation and result in expression of Tn antigen on the surface of tumor cells (Tn+ cells), which is associated with the metastasis and prognosis of cancer progression.

Oxygen by Carbon Replacement at the Glycosidic Linkage Modulates the Sugar Conformation in Tn Antigen Mimics [PDF]

open access: yes, 2018
N-Acetylgalactosamine (GalNAc) α-O-linked to l-threonine (Thr) (Tn antigen) and several mimics of this Tn antigen have been synthesized to explore the impact of the underlying amino acid in the presentation mode of the carbohydrate moiety. The structural

Expression of tumor-associated antigens in breast cancer subtypes

open access: yesBreast, 2020
Objectives: Tumor-associated antigens (TAAs) are frequently overexpressed in several cancer types. The aim of this study was to investigate the expression of TAAs in breast cancer.

An mRNA mix redirects dendritic cells towards an antiviral program, inducing anticancer cytotoxic stem cell and central memory CD8+ T cells

open access: yesFrontiers in Immunology, 2023
Dendritic cell (DC)-maturation stimuli determine the potency of these antigen-presenting cells and, therefore, the quality of the T-cell response. Here we describe that the maturation of DCs via TriMix mRNA, encoding CD40 ligand, a constitutively active ...

Expression of the Tn antigen on erythroid cells from a patient with Tn syndrome [PDF]

open access: yesJapanese journal of human genetics, 1992
In order to examine expression of the Tn antigen on erythroid cells from a patient with Tn syndrome, we applied a selective two phase liquid culture system for human erythroid progenitors in peripheral blood. The cells were analyzed with flow cytometry employing an anti-Tn antibody and a lectin of Vicia villosa which recognizes only the Tn determinant.

Efficient and Reproducible Synthesis of an Fmoc-protected Tn Antigen

open access: yes, 2021
The total synthesis of the Thomsen-nouveau (Tn) antigen, a tumour-associated O-linked mucin glycopeptide, was achieved through a concise route. The key glycosylation step proved challenging to reproduce from the literature precedents and was most ...

Emerging Roles of the Unique Molecular Chaperone Cosmc in the Regulation of Health and Disease

open access: yesBiomolecules, 2022
The core-1 β1-3galactosyltransferase-specific chaperone 1 (Cosmc) is a unique molecular chaperone of core-1 β1-3galactosyltransferase(C1GALT1), which typically functions inside the endoplasmic reticulum (ER).

Targeting Tn-positive tumors with an afucosylated recombinant anti-Tn IgG

open access: yesScientific Reports, 2023
The aberrant expression of the Tn antigen (CD175) on surface glycoproteins of human carcinomas is associated with tumorigenesis, metastasis, and poor survival.
