Nonlinearities and Switch‐Like Behavior in Gene Expression: From Genetics to Biochemistry and Back

open access: yesThe FASEB Journal, Volume 40, Issue 16, 31 August 2026.
Modest changes in transcription‐factor or chromatin‐regulator dosage can have disproportionate effects when gene‐regulatory systems operate near nonlinear thresholds. Sigmoidal input–output relationships, chromatin and cofactor recruitment, and changes in threshold position or response steepness provide molecular routes by which quantitative dosage ...

CPTCs Drive Somatic‐Visceral Communication via the Wnt Axis in Somatic Mechanotherapy: A Single‐Cell Deep Learning Study

open access: yesThe FASEB Journal, Volume 40, Issue 15, 15 August 2026.
Using an interpretable deep learning framework (CARSS) applied to multi‐tissue single‐cell RNA sequencing, we identify CD34⁺PDGFRα⁺ telocytes (CPTCs) as the primary mechanosensors bridging somatic acupuncture stimulation and visceral repair. Mechanical force at acupoints activates an AP‐1/Hsp70 immediate‐early gene program in fascial CPTCs, triggering ...

Foodborne toxin Aflatoxin B1 induced glomerular podocyte inflammation through proteolysis of RelA, downregulation of miR-9 and CXCR4/TXNIP/NLRP3 pathway

open access: yesFood Science and Human Wellness
Aflatoxin B1 (AFB1) is a naturally-occurring mycotoxin and recognized as the most toxic foodborne toxin, particularly causing damages to kidney. Glomerular podocytes are terminally differentiated epithelial cells.

Pull‐and‐Push Nanotherapeutic Hydrogels: Scavenging Inflammatory Triggers While Driving Tissue Regeneration in Burn Wounds

open access: yesAdvanced Functional Materials, Volume 36, Issue 63, 6 August 2026.
A nanounit‐assembled hydrogel employing a “pull‐and‐push” strategy simultaneously scavenges pro‐inflammatory cell‐free DNA (cfDNA) and delivers regenerative therapeutics in response to burn‐induced hyperthermia. By repolarizing macrophages and promoting angiogenesis, this multifunctional platform accelerates burn wound healing, offering a blueprint for

PRMT1‐Mediated LDHA Methylation Drives STAT3 Lactylation to Orchestrate Intestinal Inflammation and Tumorigenesis

open access: yesAdvanced Science, Volume 13, Issue 48, 28 August 2026.
This study identifies an immunometabolic axis wherein SAM‐driven PRMT1 methylates LDHA, enhancing its activity. The resultant lactate induces STAT3 K709 lactylation, which stabilizes an active conformation to promote STAT3 phosphorylation and IL‐10 expression.

Targeting Dnmt3a/m5C/RelA Axis Attenuates Microglia Inflammatory Response and Improves Postoperative Recovery in Chronic Compressive Cervical Spinal Cord Injury

open access: yesAdvanced Science, Volume 13, Issue 48, 28 August 2026.
A pro‐inflammatory microglial subset persists after decompression in chronic compressive cervical spinal cord injury and shows enhanced Dnmt3a‐driven m5C signaling. By stabilizing RelA mRNA, this axis sustains NF‐κB activation and postoperative neuroinflammation.

Kaempferol Attenuates Spaceflight‐Associated Knee Cartilage Degradation by Targeting NOX4‐Mediated Mitochondrial Dysfunction

open access: yesAdvanced Science, Volume 13, Issue 48, 28 August 2026.
We have shown that both simulated and actual spaceflight contribute to cartilage degradation in the knees of mice. Using RNA sequencing and bioenergetic profiling, we identified NADPH oxidase 4 (NOX4) as a key factor driving these changes. Additionally, we found that kaempferol, a naturally occurring flavonoid that binds directly to NOX4, reduces ...
