TSHR‐Targeting Nucleic Acid Aptamer Treats Graves' Ophthalmopathy via Novel Allosteric Inhibition

open access: yesAdvanced Science, EarlyView.
This study presents YC3, a novel inhibitory TSHR‐targeting aptamer, as a promising therapeutic for Graves' ophthalmopathy (GO). YC3 suppresses pathological phenotypes in human orbital fibroblasts and improves outcomes in GO mice by binding to a previously unidentified allosteric site on TSHR, demonstrating the potential of aptamers in advancing ...

Stochastic kinetics of ribosomes: single motor properties and collective behavior [PDF]

open access: yes, 2009
Synthesis of protein molecules in a cell are carried out by ribosomes. A ribosome can be regarded as a molecular motor which utilizes the input chemical energy to move on a messenger RNA (mRNA) track that also serves as a template for the polymerization ...

Elevation in plasma tRNA fragments precede seizures in human epilepsy.

open access: yesJournal of Clinical Investigation, 2019
Transfer RNAs (tRNAs) are a major class of noncoding RNA. Stress-induced cleavage of tRNA is highly conserved and results in tRNA fragments. Here we find specific tRNA fragments in plasma are associated with epilepsy.

tRNA‐Derived Fragment tRF‐22 Promotes Immunosuppression by Inhibiting HnRNPAB Ubiquitination in Esophageal Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
tRF‐22 fosters an immunosuppressive and pro‐oncogenic tumor microenvironment in ESCC by stabilizing hnRNPAB against TRIM25‐mediated ubiquitination and enhancing TGFβ2 expression, which increases PMN‐MDSCs infiltration and suppresses CD8+ T cells. Targeting tRF‐22 or blocking TGFβ signalling improves anti‐PD1 response, offering a promising therapeutic ...

Lactylated SPTAN1 Accelerates Hepatocellular Carcinoma Progression by Promoting NOTCH1/HES1 Activation and Immunosuppression

open access: yesAdvanced Science, EarlyView.
This study identifies SPTAN1‐kla as a novel oncogenic driver in HBV‐driven HCC, linking metabolic, epigenetic, and immune mechanisms. SPTAN1‐kla promotes HCC progression via NOTCH1/HES1 activation and immunosuppression. Findings highlight that targeting SPTAN1‐kla provides a framework for HCC treatment to improve clinical outcomes in HBV infected ...

METTL1‐Mediated M7G tRNA Modification Promotes Residual Liver Regeneration After Hepatectomy via Translational Control

open access: yesAdvanced Science, EarlyView.
METTL1 and its mediated m7G tRNA modification are significantly up‐regulated after partial hepatectomy (PHx). Overexpression of METTL1 improves the efficiency of liver regeneration after PHx, increases the proliferation of hepatocytes, and accelerates the recovery of liver function.

open access: yes, 2011
Background Transfer RNAs are synthesized as a primary transcript that is processed to produce a mature tRNA. As part of the maturation process, a subset of the nucleosides are modified.
