The RNA degradosome promotes tRNA quality control through clearance of hypomodified tRNA

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2019
Significance The cellular mechanisms governing tRNA stability in bacteria are not well understood, and in contrast to eukaryotes, a bacterial system for degradation of mature tRNAs has not been described.

AP2M1 Amplification Orchestrates Notch‐Mediated Chemoresistance in Hematopoietic Stem Cells of Acute Myeloid Leukemia Patients

open access: yesAdvanced Science, EarlyView.
The precise regulation of AP2M1 is essential for normal hematopoiesis. Overexpression of AP2M1 negatively affects the clinical outcomes of AML patients. Upregulation of AP2M1 enhances stemness and chemoresistance in HSPC cells of AML. AP2M1 modulates NOTCH1 expression, enhancing the Notch1 signaling pathway.

Multiple Layers of Stress-Induced Regulation in tRNA Biology

open access: yesLife, 2016
tRNAs are the fundamental components of the translation machinery as they deliver amino acids to the ribosomes during protein synthesis. Beyond their essential function in translation, tRNAs also function in regulating gene expression, modulating ...

Transfer RNA genes experience exceptionally elevated mutation rates. [PDF]

open access: yes, 2018
Transfer RNAs (tRNAs) are a central component for the biological synthesis of proteins, and they are among the most highly conserved and frequently transcribed genes in all living things.

N6-methyladenosine in mRNA disrupts tRNA selection and translation elongation dynamics

open access: yesNature Structural &Molecular Biology, 2016
N6-methylation of adenosine (forming m6A) is the most abundant post-transcriptional modification within the coding region of mRNA, but its role during translation remains unknown. Here, we used bulk kinetic and single-molecule methods to probe the effect

Evolution of Multivalent Aptamer Corona for High‐Throughput Multiplexed Detection of Multiple Cancers

open access: yesAdvanced Science, EarlyView.
A multivalent aptamer corona is evolved via established ProteoFish‐SELEX (systematic evolution of ligands by exponential enrichment) for high‐throughput multiplexed detection of multiple cancers. Clinical validation using multivalent aptamer corona demonstrated high accuracy and specificity in multiplexed detection of ovarian, lung, and colorectal ...

Ribosomal Synthesis of Topologically Defined Thioisoindole‐Bridged Bicyclic Peptides

open access: yesAngewandte Chemie, EarlyView.
A flexible in vitro translation (FIT) system‐based strategy for ribosomal synthesis of thioisoindole‐bridged bicyclic (TiB) peptides is developed. The flexizyme substrate Ac‐Ala(NtBA)Sc‐CME is efficiently incorporated into FIT to generate ribosomal peptides bearing N‐terminal Ac‐Ala(NtBA)Sc as the cyclization handle, which after mild deprotection ...

Impact of tRNA Modifications and tRNA-Modifying Enzymes on Proteostasis and Human Disease

open access: yesInternational Journal of Molecular Sciences, 2018
Transfer RNAs (tRNAs) are key players of protein synthesis, as they decode the genetic information organized in mRNA codons, translating them into the code of 20 amino acids. To be fully active, tRNAs undergo extensive post-transcriptional modifications,

Post-transcriptional regulation of the steady-state levels of mitochondrial tRNAs in HeLa cells [PDF]

open access: yes, 1993
In human mitochondrial DNA (mtDNA), the tRNA genes are located in three different transcription units that are transcribed at three different rates. To analyze the regulation of tRNA formation by the three transcription units, we have examined the steady-
