Wet Granulation in a Twin‐Screw Extruder: Implications of Screw Design

Journal of Pharmaceutical Sciences, 2010
Wet granulation in twin-screw extrusion machinery is an attractive technology for the continuous processing of pharmaceuticals. The performance of this machinery is integrally tied to its screw design yet little fundamental knowledge exists in this emerging field for granulation to intelligently create, troubleshoot, and scale-up such processes.

Twin screw wet granulation: The study of a continuous twin screw granulator using Positron Emission Particle Tracking (PEPT) technique

European Journal of Pharmaceutics and Biopharmaceutics, 2012
In this paper, Positron Emission Particle Tracking (PEPT) techniques are utilised to track the trajectory of single particles through the mixing and conveying zones of a Twin Screw Granulator (TSG). A TSG consisting of conveying zones and mixing zones is used in this study. The mixing zones are arranged with kneading discs at an angle of 30°, 60° or 90°

Twin screw granulation – review of current progress

Drug Development and Industrial Pharmacy, 2014
Twin screw granulation (TSG) is a new process of interest to the pharmaceutical community that can continuously wet granulate powders, doing so at lower liquid concentrations and with better product consistency than found by a high shear batch mixer. A considerable body of research has evolved over the short time since this process was introduced but ...

Dissolution enhancement of carbamazepine using twin-screw melt granulation

European Journal of Pharmaceutics and Biopharmaceutics, 2020
The current study explored the twin-screw melt granulation (TSMG) as a potential technology for the water solubility enhancement of biopharmaceutical classification system (BCS) class II drugs. As a model drug, carbamazepine (CBZ) was formulated with three different polymers as melt granules produced in a co-rotating twin-screw granulator. Polyethylene

Twin screw wet granulation: Effects of properties of granulation liquid

Powder Technology, 2012
Abstract The effect of the properties of the granulation liquid, i.e. viscosity (amount of binder) and surface tension on residence time, torque and granule properties such as size, shape, structure, strength and flowability was studied in a co-rotating twin screw granulator.

Comparison of two twin-screw extruders for continuous granulation

European Journal of Pharmaceutics and Biopharmaceutics, 2009
A comparison was made between two twin-screw extruders (APV Baker and Leistritz Micro) used for continuous wet granulation. Both extruders had similar screw configurations, based on the length-to-diameter ratio of the screws, existing out of a conveying zone of 20 D, i.e. 20 times the screw diameter and a granulation zone of 4D.

Influence of filler selection on twin screw foam granulation

Drug Development and Industrial Pharmacy, 2013
The influence of filler selection in wet granulation was studied for the novel case where the binder is delivered as an unstable, semi-rigid aqueous foam to an extrusion process. The work primarily examined the impact of differing concentrations of microcrystalline cellulose (Avicel PH® 101) in a formulation with spray-dried α-lactose monohydrate ...

Melt granulation: A comparison of granules produced via high-shear mixing and twin-screw granulation

International Journal of Pharmaceutics, 2020
Melt granules of DI-CAFOS® A12 and 15% (w/w) Kolliphor® P407 were manufactured in a twin-screw granulator (TSG) at five different conditions (screw speed and throughput varied) and compared to granules manufactured in a high-shear granulator (HSG) (rotation speed of chopper/impeller and granulation time varied).

Impact of screw elements on continuous granulation with a twin-screw extruder

Journal of Pharmaceutical Sciences, 2008
The influence of different screw element types on wet granulation process with a twin-screw extruder was investigated. Lactose granules were prepared with different screw configurations such as conveying, combing mixer and kneading elements. The use of kneading blocks led to an almost complete agglomeration of lactose, whereas kneading and combing ...

Influence of granulation temperature on particle size distribution of granules in twin-screw granulation (TSG)

Pharmaceutical Development and Technology, 2019
This study investigated an influence of granulation temperature during twin-screw granulation (TSG) on particle size distributions (PSDs). The influence of the granulation temperature on granule size distributions varied, depending on the liquid to solid (L/S) ratio, the kind of binders, the method of binder addition, and the filler material.
