Preclinical validation of a clinical prototype of intraoperative electron FLASH radiotherapy equipment: key evidence for a new radiotherapy paradigm

open access: yesRadiation Oncology
Background and purpose This study aimed to evaluate whether the clinical prototype of intraoperative electron FLASH radiotherapy (CPIO-EFLASH), with a source-surface distance (SSD) of 50 cm, could achieve ultra-high dose rates, effectively control tumors,

A Biomimetic cRGDyk‐Engineered Platelet Nanoplatform for ACAD11 Delivery to Trigger Pyroptosis in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
A biomimetic platelet nanoplatform (OE‐ACAD11@R‐PLT) is developed for targeted hepatocellular carcinoma immunotherapy. By selectively delivering therapeutic plasmids, this system induces mitochondrial dysfunction and triggers robust NLRP3/Caspase‐1/GSDMD‐mediated pyroptosis in tumor cells.

Advancing Biotherapeutic Treatment of Ulcerative Colitis Using Lipid Mesophases: Formulation Design and Preclinical Validation

open access: yesAdvanced Science, EarlyView.
Lipid mesophases provide a versatile platform for colon‐targeted delivery of biotherapeutics, protecting them from degradation while enhancing mucoadhesion and enabling localized, controlled release to the inflamed mucosa after oral or rectal administration. In a DSS‐induced colitis model, this strategy reduced disease activity, inflammatory biomarkers,

MOSkin dosimetry for an ultra-high dose-rate, very high-energy electron irradiation environment at PEER

open access: yesFrontiers in Physics
FLASH radiotherapy, which refers to the delivery of radiation at ultra-high dose-rates (UHDRs), has been demonstrated with various forms of radiation and is the subject of intense research and development recently, including the use of very high-energy ...

Fc‐Dependent Targeting of the GAS6–MERTK Axis With Monoclonal and Bispecific Antibodies Overcomes Drug Resistance Across EGFR Mutations in Exons 19, 20, and 21

open access: yesAdvanced Science, EarlyView.
EGFR mutations drive a substantial fraction of non‐small cell lung cancers (NSCLC) and are effectively targeted by tyrosine kinase inhibitors (TKIs), including osimertinib. However, resistance inevitably develops through secondary mutations and compensatory receptor activation, including the GAS6 receptor MERTK.

ZMIZ2–DUX4–LGR5 Signaling Axis Maintains Stemness Properties and Facilitates Docetaxel Resistance in Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
This study identifies ZMIZ2 as a therapeutic target for docetaxel‐resistant triple‐negative breast cancer, sustaining cancer stemness‐associated properties and docetaxel resistance. ZMIZ2 upregulation enhances DUX4‐mediated LGR5 activation through the SWI/SNF components BRG1 and BAF57.

Photoactivated Platinum Complexes: Targeting Membrane Proteins and Organelles for Precision Cancer Therapy

open access: yesAngewandte Chemie, EarlyView.
This review highlights an emerging anticancer approach integrating platinum agents into photodynamic therapy (PDT) and photoactivated chemotherapy (PACT) systems, further augmented by cancer cell membrane‐ and organelle‐targeting. By directing photoactivated platinum complexes from nontargeted distribution to cancer cells or defined subcellular sites ...

NSUN3‐Associated m5C Modification of tRF5‐GlyTCC Suppresses Colorectal Cancer Progression via RPL23‐Linked Nucleolar Stress and p53‐Associated Autophagy and Enhances 5‐FU Sensitivity

open access: yesAdvanced Science, EarlyView.
This study identifies that NSUN3‐mediated m5C modification stabilizes tRF5‐GlyTCC, linking ribosomal homeostasis, nucleolar stress responses, and p53 signaling changes. This regulatory axis suppresses colorectal cancer and enhances chemotherapy response to 5‐FU, positioning tRF5‐GlyTCC as a potential therapeutic target for improving drug sensitivity in
