Human VNTR mutation and sex

Seven hypervariable VNTR loci have been studied in about 1200 parent/child pairs about equally divided between the sexes. Mutations were observed with all seven probes, the total number being 71. Fourty-four of these involved increased fragment length. Gains in fragment length were on average larger than losses.

By cloning tandemly repeated sequences from the pig genome by use of non-porcine minisatellite probes for library screening, five novel polymorphic VNTR loci were isolated: three minisatellites and two satellite-like loci. Four of them could be mapped onto chromosomes by linkage analysis and/or in situ hybridization.

Mapping of complex genomes has been influenced substantially by the isolation of locus-specific, but repetitive DNA elements known as VNTRs. Since a high GC-content is characteristic of most of these elements one might expect them to be clustered at least partially in CpG-islands.
