Electrostatic charging artefacts in Lorentz electron tomography of MFM tip stray fields [PDF]

open access: yes, 2001
Using the technique of differential phase contrast (DPC) Lorentz electron microscopy, the magnetic stray field distribution from magnetic force microscopy (MFM) tips can be calculated in a plane in front of the tip using tomographic reconstruction ...

Protocol for quantifying miRNA trafficking across the endosomal membrane

open access: yesFEBS Open Bio, EarlyView.
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.

Total Sample Conditioning and Preparation of Nanoliter Volumes for Electron Microscopy [PDF]

open access: yesACS Nano, 2016
Electron microscopy (EM) entered a new era with the emergence of direct electron detectors and new nanocrystal electron diffraction methods. However, sample preparation techniques have not progressed and still suffer from extensive blotting steps leading to a massive loss of sample. Here, we present a simple but versatile method for the almost lossless

Transmission electron microscopy with in situ ion irradiation [PDF]

open access: yes, 2015
The macroscopic properties of materials exposed to irradiation are determined by radiation damage effects which occur on the nanoscale. These phenomena are complex dynamic processes in which many competing mechanisms contribute to the evolution of the ...

The C‐terminal domain of yeast Arginyltransferase1 is essential for its catalytic activity

open access: yesFEBS Open Bio, EarlyView.
Arginyltransferase 1 (Ate1), a eukaryotic enzyme, catalyses arginylation, transferring arginine from tRNA‐Arg to the amino terminus of the target protein. Overexpression of Ate1 in yeast is lethal and is dependent on arginylation. This study elucidates how mutations in the cofactor‐binding and active site of Ate1 and truncation of its structural ...

Imaging and 3D reconstruction of membrane protein complexes by cryo-electron microscopy and single particle analysis [PDF]

open access: yes, 2006
Cryo-electron microscopy (cryo-EM) in combination with single particle image processing and volume reconstruction is a powerful technology to obtain medium-resolution structures of large protein complexes, which are extremely difficult to crystallize and ...

Dactolisib outperforms Rapamycin in potentiating apoptosis induced by XPO1, Bcl‐2 and Mcl‐1 blockade in preclinical glioblastoma models

open access: yesFEBS Open Bio, EarlyView.
The PAM pathway and its impact on cellular metabolism. Color‐coded key symbols represent Rapamycin (R, orange, targeting mTOR) and Dactolisib (D, green, targeting PI3K/mTOR), indicating their sites of action. Created with BioRenders.com. Glioblastoma resists therapy through interconnected survival programs.

A cross-sectional transmission electron microscopy study of iron recovered from a laser-heated diamond anvil cell [PDF]

open access: yes, 2008
This paper discusses the use of a focused ion beam for the preparation of cross-sectional transmission electron microscopy specimens from small samples, typically 150 micron in diameter, that have been recovered from a laser-heated diamond anvil cell. We
