Opposing roles of DGAT‐mediated lipid droplet biogenesis in the regulation of ferroptosis sensitivity

Ferroptosis is driven by unrestricted peroxidation of polyunsaturated fatty acyl (PUFA) chains in membrane phospholipids. In triple‐negative breast cancer cells, lipid droplets (LDs) sequester dietary PUFAs, limiting their incorporation into membrane phospholipids, preventing mitochondrial damage, and reducing lipid peroxidation and ferroptosis.

Resistance‐exercise‐induced stress intervenes in TGF‐β signaling by cooperatively downregulating nuclear αB‐crystallin and SMAD4 in human skeletal muscle fibers

Alpha‐crystallin B chain (CRYAB) has been reported to stabilize mothers against decapentaplegic homolog 4 (SMAD4) in transforming growth factor‐β (TGF‐β) signaling, enabling target gene transcription. We show nuclear CRYAB–SMAD4 interaction for the first time in human skeletal muscle fibers and its regulation by exercise.

Quantitative analysis of DNA‐GATA1 binding alterations linked to hematopoietic disorders

Native holdup allows the quantitative determination of affinities between full‐length transcription factors and DNA. Mutations in either the protein or the DNA can modulate binding strength, which can be precisely quantified using this approach. Applied to GATA1, it revealed mutations that alter DNA binding.
