Engineering antibodies by yeast display
Archives of Biochemistry and Biophysics, 2012Since its first application to antibody engineering 15 years ago, yeast display technology has been developed into a highly potent tool for both affinity maturing lead molecules and isolating novel antibodies and antibody-like species. Robust approaches to the creation of diversity, construction of yeast libraries, and library screening or selection ...

Yeast cell-surface display?applications of molecular display
Applied Microbiology and Biotechnology, 2004In a cell-surface engineering system established using the yeast Saccharomyces cerevisiae, novel, so-called arming yeasts are constructed that are armed with biocatalysts in the form of enzymes, functional proteins, antibodies, and combinatorial protein libraries.

Protein selection using yeast surface display
Methods, 2013Binding proteins are typically isolated from combinatorial libraries of scaffold proteins using one of the many library screening tools available, such as phage display, yeast surface display or mRNA display. A key principle underlying these screening technologies is the establishment of a link between each unique mutant protein and its corresponding ...

Quantification analysis of yeast-displayed lipase
Analytical Biochemistry, 2014We describe a method for quantification of displayed lipase on yeast cell surface. The strategy uses an organophosphonate ester to irreversibly inhibit the active lipase and release a detectable fluorescent group. The amount of displayed lipase can be represented as "g/g cell" or "molecules/cell".

Protein Engineering by Yeast Surface Display
Journal of Visualized ExperimentsProtein engineering enables the improvement of existing functions of a given protein or the generation of novel functions. One of the most widely used and versatile tools in the protein engineering field is yeast surface display, where a pool of randomized proteins is expressed on the surface of yeast.

Optimized protocol for antibody yeast display
2023Yeast display is an established technology used to discover proteins that have increased binding affinities to their targets. There are many steps within the process that affect the final binding affinity between proteins. The Maynard Lab regularly utilizes display technologies to discover antibodies that bind strongly to targets of interest prior to ...

Engineering Antibody Affinity by Yeast Surface Display
2004Publisher Summary This chapter elaborates the engineering antibody affinity by yeast surface display (YSD). YSD is a powerful tool for engineering the affinity, specificity, and stability of antibodies, as well as other proteins. The methods for displaying an antibody on yeast, creating mutant libraries and sorting libraries for improved clones are ...

Simultaneous Yeast Display for Medical Biosensors
2023Biosensors play an important role in medical diagnostics, industrial biotechnology, and environmental monitoring. A key challenge in biosensor design is developing complementary ligand binding domains which bind to the small molecule in a sandwich like fashion, causing fused biosensor output domains to co-localise and transduce a signal.
