Isolating and engineering human antibodies using yeast surface display
Nature Protocols, 2006This protocol describes the process of isolating and engineering antibodies or proteins for increased affinity and stability using yeast surface display. Single-chain antibody fragments (scFvs) are first isolated from an existing nonimmune human library displayed on the yeast surface using magnetic-activated cell sorting selection followed by selection

Epitope-Specific Binder Design by Yeast Surface Display
2015Yeast surface display is commonly used to engineer affinity and design novel molecular interaction. By alternating positive and negative selections, yeast display can be used to engineer binders that specifically interact with the target protein at a defined site.

Yeast surface display of dehydrogenases in microbial fuel-cells
Bioelectrochemistry, 2016Two dehydrogenases, cellobiose dehydrogenase from Corynascus thermophilus and pyranose dehydrogenase from Agaricus meleagris, were displayed for the first time on the surface of Saccharomyces cerevisiae using the yeast surface display system. Surface displayed dehydrogenases were used in a microbial fuel cell and generated high power outputs.

Antibody Affinity Optimization Using Yeast Cell Surface Display
2009Many biosensors depend on molecular recognition reagents to achieve highly sensitive and specific detection levels of an analyte of interest. Although new and improved detection platforms continue to be developed, improvements in the affinity and specificity of the molecular recognition reagents often dictate the ultimate performance level and utility ...

Engineering Fibronectin-Based Binding Proteins by Yeast Surface Display
2013Yeast surface display (YSD) presents proteins on the surface of yeast through interaction of the agglutinin subunits Aga1p and Aga2p. The human 10th type III fibronectin (Fn3) is a small, 10-kDa protein domain that maintains its native fold without disulfide bonds.

The Use of Yeast Surface Display in Biofuel Cells
2015Biofuel cells are electrochemical devices which convert chemical energy to electricity using biochemical pathways and redox enzymes. In enzymatic fuel cells purified redox enzymes catalyze the reactions in the anode and cathode compartments whereas in microbial fuel cells (MFCs) the entire metabolism of the microorganisms is exploited.

A facile yeast-display approach for antibody mask discovery
Protein Engineering, Design and SelectionAbstract Tuning in vivo activity of protein therapeutics can improve their safety. In this vein, it is possible to add a ‘mask’ moiety to a protein therapeutic such that its ability to bind its target is prevented until the mask has been proteolytically removed, for instance by a tumor-associated protease. As such, new methods to isolate

Surface display of proteins in yeasts
2014Surface display of proteins in bacteria and yeasts has proved to be an interesting method of protein immobilization for different purposes. Generally, proteins of interest are genetically engineered by fusing them to autochthonous cell wall proteins, or parts of autochthonous proteins directing them to the wall and anchoring them at the cell surface ...
