Simplified Switching to Once‐Weekly Insulin Icodec Without an Initial One‐Time Additional Dose Versus Once‐Daily Insulin Glargine U100 in Basal Insulin‐Treated Type 2 Diabetes (ONWARDS 10): A Randomised Controlled Trial

open access: yesDiabetes, Obesity and Metabolism, EarlyView.
ABSTRACT Aims ONWARDS 10 assessed switching from daily basal insulins to once‐weekly insulin icodec without a one‐time additional dose at first injection. Materials and Methods In this treat‐to‐target, open‐label, 26‐week, phase 3b trial, adults with basal‐insulin‐treated type 2 diabetes (T2D) with glycated haemoglobin [HbA1c] 7.0%–10.0% were ...

Glucagon‐Like Peptide‐1 Receptor Agonists and Incident Major Adverse Liver Outcomes in People With Type 2 Diabetes and Metabolic Dysfunction‐Associated Steatotic Liver Disease

open access: yesDiabetes, Obesity and Metabolism, EarlyView.
ABSTRACT Aims Treatment options for metabolic dysfunction‐associated steatotic liver disease (MASLD) are limited. While glucagon‐like peptide‐1 receptor agonists (GLP‐1 RA) and sodium‐glucose cotransporter‐2 (SGLT‐2) inhibitors improve cardiovascular outcomes, comparative effectiveness on liver‐related outcomes remains unclear.

Design, synthesis and in vitro biological evaluation of new coumarin containing oxazole hybrid derivatives as multitarget inhibitor of α-amylase and α-glucosidase for the treatment of diabetes

open access: yesResults in Chemistry
Sixteen new coumarin-based oxazole derivatives were synthesized and their structures were characterized by employing various spectroscopic tools including HREI-MS and NMR. The in vitro assessment of their α-amylase and α-glucosidase inhibitory activities

Ubiquitin and ubiquitin‐like modifications in the endoplasmic reticulum stress response

open access: yesThe FEBS Journal, EarlyView.
Endoplasmic reticulum (ER) stress activates various proteostasis control processes, including the unfolded protein response, ribosome‐associated quality control, and ER‐associated degradation. Ubiquitin and ubiquitin‐like modifications dynamically regulate these processes to determine cell fate, promoting adaptation or inducing cell death.

Transcriptomic and proteomic signatures underlying nymphal adaptation and foam production in the forage pest Mahanarva spectabilis

open access: yesInsect Molecular Biology, EarlyView.
RNA‐seq and Batelli gland proteomics of fifth‐instar Mahanarva spectabilis nymphs reveal transcripts and proteins associated with xylem feeding, foam production and environmental interaction. Functional annotation identified genes involved in osmoregulation, detoxification, chemosensation and stress responses, while proteomic analysis confirmed ...

Neutrophil Secretory Proteins Inhibit Calcium Oxalate Crystallisation and Crystal Growth, but Promote Crystal Aggregation

open access: yesImmunology, EarlyView.
Calcium oxalate monohydrate (COM) crystals cause changes in the secretion of 29 proteins from neutrophil‐like cells, including those involved in neutrophil immune activation and intracellular metabolic adaptation. The neutrophil secreted proteins (secretome) also inhibit calcium oxalate crystallisation and crystal growth, but promote crystal ...

Primary cardiovascular preventive effect of thiazolidinedione in adults with type 2 diabetes: A nationwide cohort study in Korea

open access: yesJournal of Diabetes Investigation, EarlyView.
Among Korean adults with type 2 diabetes and no prior cardiovascular disease, thiazolidinedione use was associated with a lower risk of the primary composite outcome of stroke, myocardial infarction, hospitalization for heart failure, and all‐cause mortality compared to dipeptidyl peptidase‐4 inhibitor use. ABSTRACT Background Thiazolidinediones (TZDs)

Risk of urinary tract infection with SGLT2 inhibitor initiation in patients with immune‐mediated inflammatory diseases and type 2 diabetes: A target trial emulation using a Japanese hospital‐based claims database

open access: yesJournal of Diabetes Investigation, EarlyView.
In this target trial emulation of patients with immune‐mediated inflammatory diseases and type 2 diabetes, SGLT2 inhibitor initiation was not associated with a clear increase in UTI risk versus metformin, including across immunosuppressive therapy strata.

Dapagliflozin does not impair microvascular hemorheology in type 2 diabetes mellitus: A multicenter randomized controlled trial (D‐PATH study)

open access: yesJournal of Diabetes Investigation, EarlyView.
Graphical abstract illustrating the study design, physiological rationale, and key findings of the D‐PATH trial. Dapagliflozin, an SGLT2 inhibitor, lowers HbA1c and body weight while increasing hematocrit and erythropoietin levels. Although an elevated hematocrit could theoretically increase blood viscosity and impair microcirculatory flow, ex vivo ...
