Highlights Diabetic kidney disease is a progressive disease with the pathology of glomerular damage and loss of podocytes. Sodium glucose cotransporter 2 inhibitors (SGLT2i) can be expected to decrease the amount of podocyte excretion. 目的 通过基因沉默技术构建质粒成功转染原代星形胶质细胞(Ast)抑制IL-17表达,探讨IL-17的功能。方法 在制备原代Ast基础上,应用IL-17基因沉默质粒pGenesil-1A和B,用Lipofectamine 2000转染Ast,通过荧光分析法、RT-PCR法、Western Blotting法观察IL-17基因沉默后效果。结果 携带pGenesil-1B和A的IL-17基因沉默质粒制备成功后,免疫荧光携带pGenesil-1B转染效果明显高于pGenesil-1A,其转染率可高达56.72%;
