目的 构建带有FLAG标签的BPIFC基因真核表达载体,建立稳定转染BPIFC基因的HaCaT细胞系。方法 设计引物通过PCR扩增出BPIFC基因片段,利用DNA重组技术将该基因片段定向插入pcDNA3.1-flag-N真核表达载体中,并对重组质粒进行双酶切及DNA测序鉴定。验证正确的重组质粒通过PI Max线性转染的方法转染至HaCaT细胞,通过G418筛选建立BPIFC稳定转染的HaCaT细胞系,并通过RT-PCR法和Western-blot法分别测定BPIFC mRNA和蛋白的表达量。结果 ...
