ADAMTS4 is involved in the production of the Alzheimer disease amyloid biomarker APP669-711

open access: yesMolecular Psychiatry, 2023
AbstractAmyloid-β (Aβ) deposition in the brain parenchyma is one of the pathological hallmarks of Alzheimer disease (AD). We have previously identified amyloid precursor protein (APP)669-711 (a.k.a. Aβ(-3)-40) in human plasma using immunoprecipitation combined with matrix-assisted laser desorption ionization time-of-flight mass spectrometry (IP-MALDI ...

Subregion‐specific perineuronal net remodelling in the dorsal hippocampus drives vulnerability and resilience to chronic stress: The role of venlafaxine treatment

open access: yesBritish Journal of Pharmacology, EarlyView.
Background and Purpose Neuronal plasticity enables the brain to adapt to internal and external demands by dynamically regulating synaptic connectivity and emotional circuitry. Impaired neuroplasticity represents a core pathophysiological framework of major depressive disorder, one of the leading causes of disability worldwide. Chronic stress is a major

Crystal structures of the two major aggrecan degrading enzymes, ADAMTS4 and ADAMTS5 [PDF]

open access: yesProtein Science, 2008
AbstractAggrecanases are now believed to be the principal proteinases responsible for aggrecan degradation in osteoarthritis. Given their potential as a drug target, we solved crystal structures of the two most active human aggrecanase isoforms, ADAMTS4 and ADAMTS5, each in complex with bound inhibitor and one wherein the enzyme is in apo form.

Brevican Is Degraded by Matrix Metalloproteinases and Aggrecanase-1 (ADAMTS4) at Different Sites [PDF]

open access: yesJournal of Biological Chemistry, 2000
Brevican is a member of the lectican family of chondroitin sulfate proteoglycans that is predominantly expressed in the central nervous system. The susceptibility of brevican to digestion by matrix metalloproteinases (MMP-1, -2, -3, -7, -8, -9, -10, and -13 and membrane type 1 and 3 MMPs) and aggrecanase-1 (ADAMTS4) was examined. MMP-1, -2, -3, -7, -8,

ADAMTS4 mediates LPS‐induced cardiac injury and myocardial fibrosis through proteolytic cleavage of TSP1

open access: yesClinical and Translational Medicine, Volume 16, Issue 8, August 2026.
• ADAMTS4 is induced primarily in cardiac fibroblasts during sepsis. • ADAMTS4 cleaves TSP1 at the 236–246 aa functional region. • TSP1 cleavage activates TGF‐β/Smad and NF‐κB signalling in fibroblasts. • Targeting ADAMTS4 reduces fibroblast activation and cardiac fibrosis.

Divergent Atrial Transcriptomic Signatures in Diabetic and Nondiabetic Atrial Fibrillation

open access: yesJournal of Arrhythmia, Volume 42, Issue 4, August 2026.
Diabetes alters the atrial transcriptome in atrial fibrillation, characterized by enhanced mitochondrial ATP synthesis, suppressed immune and inflammatory pathways, and distinct miRNA expression profiles, suggesting metabolic and epigenetic remodeling of the atrial substrate.

Drug‐Induced Cuproptosis Defines the Therapeutic Window of Celecoxib in Intervertebral Disc Degeneration via the HSP90‐RBX1 Axis

open access: yesAdvanced Science, Volume 13, Issue 42, 28 July 2026.
This study reveals the dual role of celecoxib in intervertebral disc degeneration. While low concentrations are protective, high concentrations induce toxicity by upregulating HSP90, which synergizes with USP15 to deubiquitinate and stabilize RBX1. This leads to degradation of COMMD1/ATP7B, copper dyshomeostasis, and ultimately, cuproptosis.
