Comprehensive Investigation of Benzimidazolium Derivatives Through Synthesis, Characterization, Cytotoxicity Activity, Immunocytochemistry, and ADME and Toxicity Analyses

open access: yesChemistryOpen, Volume 15, Issue 9, September 2026.
Three benzimidazolium derivatives bearing fluoro, chloro, and methoxy substituents are synthesized and evaluated for anticancer activity. Compound 2b shows the highest antiproliferative activity and induces apoptosis through caspase‐3 activation and PARP1 cleavage. In this study, three novel benzimidazolium derivatives (2a–c) bearing 4‐fluoro, 3‐chloro,

Integrated In Silico Discovery of Thymoquinone Analogs Targeting the Keap1–Nrf2 Pathway for Amyotrophic Lateral Sclerosis Therapy

open access: yesChemistryOpen, Volume 15, Issue 9, September 2026.
Integrated in silico discovery of thymoquinone analogs as potent Keap1 inhibitors for ALS therapy. Using validated QSAR, docking, and 200 ns MD simulations, we identified brain‐penetrant leads (CHEMBL3416163, CHEMBL4636830, and CHEMBL221598) with binding affinities up to fourfold stronger than thymoquinone, offering a targeted strategy to restore redox

Synthesis and Biological Activity of 2,6‐Disubstituted and 2,6,9‐Trisubstituted 7‐Deazapurine Nucleobases

open access: yesChemMedChem, Volume 21, Issue 16, 27 August 2026.
Four series of 20 (het)aryl substituted 7‐deazapurine nucleobases were synthesized. Several potent antagonists of all four subtypes of adenosine receptors were identified by functional assay. High‐throughput screening of the IMTM compound library for modulatory activity on adenosine receptors (ARs) identified two deazapurine hits based on 2,6‐diaryl‐7 ...

In the peculiar manner by which physicists reckon descent, this article is by a "child" and "grandchild" of the late Stanley Deser. We begin by sharing reminiscences of Stanley from over 40 years. Then we turn to a problem which was dear to his heart: the prospect that gravity might nonperturbatively screen its own ultraviolet divergences and those of ...

Rational Design of Multifunctional Tacrine Derivatives as Candidates for the Treatment of Alzheimer and Parkinson Diseases

open access: yesChemMedChem, Volume 21, Issue 15, 14 August 2026.
We present a set of multifunctional tacrine derivatives, designed with the CADMA‐Chem protocol, aimed for Alzheimer and Parkinson diseases. Of the tacrines designed, TAC‐674 and TAC‐678 are the best candidates to act as multifunctional antioxidants and neuroprotectors for Alzheimer and Parkinson diseases, respectively.
