RNA polymerase II (Pol II) is the central enzyme that transcribes eukaryotic protein-coding genes to produce mRNA. The mushroom toxin α-amanitin binds Pol II and inhibits transcription at the step of RNA chain elongation.

利用加端PCR技术在 pGEX 1λT质粒BamHI和PstI位点间 (片段BamHI端 )加入α amanitin线性八肽碳骨架编码序列 ,再将两次连续加端PCR产物连入BamHI、PstI双酶切的pGEX 1λT载体 ,得到含α amanitin线性碳骨架编码序列的阳性重组子pGAT 1 .序列测定和结果分析显示 ,构建的融合基因相位正确 ,阅读框通读 .通过对融合基因在大肠杆菌中表达条件的优化 ,目的融合蛋白的表达量可提高 33.5 %,占总蛋白表达量的 2 8.3%.
