Hypermutation in antibody affinity maturation
Current Opinion in Immunology, 1999By studying the role of mismatch repair in hypermutation at the immunoglobulin loci, the field of antibody hypermutation has been integrated into the larger area of DNA repair. Trans-acting factors - Ku70, Ku80 and possibly SWAP-70 - have been identified for the temporally related but not mechanistically related immunoglobulin heavy-chain class-switch.

Peptide Affinity Purification of Antibodies
Cold Spring Harbor Protocols, 2019This protocol describes antibody purification using a peptide affinity column. Peptides can be designed that use naturally occurring cysteines within the protein target's primary sequence, or a cysteine can be added to either end of the peptide to provide free thiols for attachment.

Engineering antibody affinity and specificity
International Journal of Cancer, 1988A combination of ab initio calculations, "knowledge-based prediction", molecular graphics and site-directed mutagenesis has enabled us to probe the molecular details of antibody:antigen recognition and binding and to alter the affinity and specificity of an antibody for its antigen.

Affinity Purification of Antibodies
2015Antibodies are provided in a variety of formats that include antiserum, hybridoma culture supernatant, or ascites. They can all be used successfully in crude form for the detection of target antigens by immunoassay. However, it is advantageous to use purified antibody in defined quantity to facilitate assay reproducibility, economy, and reduced ...

Affinity-bound antibody–drug conjugates
Nature Biomedical Engineering, 2019Computationally designed antibody–drug conjugates easily self-assemble via affinity ligands that non-covalently link a therapeutic drug to conserved sites on native IgG antibodies.

Antibody affinity measurements
1997Abstract In order to assess the biological activity of antibody (Ab) molecules, it is necessary to measure the strength of their interaction with the antigen (Ag) in terms of the equilibrium constant KA (1). This constant corresponds to the inverse of the Ab concentration which is needed to obtain 50% of complexed species.

Affinity Chromatography for Antibody Purification
2014The availability of purified antibodies is prerequisite for many applications and the appropriate choice(s) of antibody-purification steps is crucial. Numerous methods have been developed for the purification of antibodies; however, affinity chromatography-based methods are the most extensively utilized.

Affinity screening for weak monoclonal antibodies
Journal of Immunological Methods, 1998When selecting for monoclonal antibodies of a desired affinity, affinity chromatography can be a feasible alternative. This is of particular interest when low affinity monoclonal antibodies (dissociation constant (Kd) > 10(-4) M) are screened, as they are not easily recognised by traditional immunoassay procedures.

Affinity chromatographic purification of morphine antibody
Life Sciences, 1976Abstract Morphine-6-hemisuccinate was synthesized and linked to agarose affinity beads by either direct amide bond formation or by an N-hydroxysuccinimide ester intermediate using various conditions. The various preparative routes resulted in differing ampunts of covalently bound ligand.

Purification of antibodies by affinity chromatography
Journal of Biochemical and Biophysical Methods, 2002This review focusses on affinity purification of immunoglobulins, a methodology which is a powerful tool to obtain pure and intact antibodies. Affinity techniques allow antibody purification both in a single step chromatographic procedure as well as in complex purification protocols depending on the intention to use the target antibody.
