A Decoy‐Receptor‐Armed Biomimetic Nanotherapeutic With Inherent Tropism for Conserved Pathogenic Macrophages for Treating Osteoarthritis and Intervertebral Disc Degeneration

open access: yesAdvanced Science, EarlyView.
A biomimetic doppelgänger nanosystem neutralizes extracellular inflammatory cytokines and silences intracellular pyroptosis, reprogramming pathogenic macrophages to attenuate both joint and spine degeneration. ABSTRACT Osteoarthritis (OA) and intervertebral disc degeneration (IVDD) are debilitating musculoskeletal disorders driven by shared ...

The role and control of arginine levels in arginase 1 deficiency

open access: yesJournal of Inherited Metabolic Disease, 2022
AbstractArginase 1 Deficiency (ARG1‐D) is a rare urea cycle disorder that results in persistent hyperargininemia and a distinct, progressive neurologic phenotype involving developmental delay, intellectual disability, and spasticity, predominantly affecting the lower limbs and leading to mobility impairment.

Decreased systemic bioavailability of L-arginine in patients with cystic fibrosis

open access: yesRespiratory Research, 2006
Background L-arginine is the common substrate for nitric oxide synthases and arginases. Increased arginase levels in the blood of patients with cystic fibrosis may result in L-arginine deficiency and thereby contribute to low airway nitric oxide ...

EBV‐Driven ODC1 Upregulation Enhances Polyamine Anabolism to Promote Viral Replication and Cisplatin Resistance in Nasopharyngeal Carcinoma

open access: yesAdvanced Science, EarlyView.
EBV‑BZLF1 initiates ODC1‑driven polyamine anabolism that correlates with poor clinical prognosis in nasopharyngeal carcinoma. The ODC1–spermidine axis promotes viral replication, cell proliferation and innate immune evasion. Pharmacological inhibition of ODC1 rescues cisplatin sensitivity, establishing ODC1 as a viable therapeutic target for EBV ...

Generation of a Mouse Model for Arginase II Deficiency by Targeted Disruption of the Arginase II Gene [PDF]

open access: yesMolecular and Cellular Biology, 2001
Mammals express two isoforms of arginase, designated types I and II. Arginase I is a component of the urea cycle, and inherited defects in arginase I have deleterious consequences in humans. In contrast, the physiologic role of arginase II has not been defined, and no deficiencies in arginase II have been identified in humans. Mice with a disruption in

Review of arginase as a promising biocatalyst: characteristics, preparation, applications and future challenges

open access: yes, 2021
As a committed step in the urea cycle, arginase cleaves l-arginine to form l-ornithine and urea. l-Ornithine is essential to: cell proliferation, collagen formation and other physiological functions, while the urea cycle itself converts highly toxic ...

ROS‐Responsive Eye Drop for Controlled Release of 20‐Deoxyingenol Improves Diabetic Corneal Healing by Activating Macrophage Autophagy and Metabolic Reprogramming

open access: yesAdvanced Science, EarlyView.
Schematic illustration of ROS‐responsive TSPBA‐PVA microspheres loaded with 20‐deoxyingenol (TP/20‐DOI) for diabetic corneal wound healing: (a) fabrication and ROS‐triggered release; (b) autophagy activation and metabolic reprogramming in macrophages; (c) suppression of macrophage‐derived CXCL3/CXCL5, attenuation of neutrophil infiltration, and ...

The role of the host—Neutrophil biology

open access: yesPeriodontology 2000, EarlyView., 2023
Abstract Neutrophilic polymorphonuclear leukocytes (neutrophils) are myeloid cells packed with lysosomal granules (hence also called granulocytes) that contain a formidable antimicrobial arsenal. They are terminally differentiated cells that play a critical role in acute and chronic inflammation, as well as in the resolution of inflammation and wound ...

Anesthetic Management Of A Pediatric Patient With Arginase Deficiency

open access: yesMedical Journal of Trakya University, 2009
Arginase deficiency is an autosomal recessive disorder of the urea cycle in which a defect in conversion of arginine to urea and ornithine leads to hyperammonemia. Patients with urea cycle disorders may show increased protein catabolism due to inadequate intake of energy, protein and essential amino acids; infections, fever and surgery.

TH17–Associated Polyamine Metabolism‐Guided Nanozyme Promotes Alveolar Bone Repair in Periodontitis

open access: yesAdvanced Science, EarlyView.
A pronounced TH17‐skewed immune response is identified in experimental periodontitis, and enrichment of the polyamine pathway is observed during TH17 differentiation, suggesting its potential as an immunometabolic target. Inspired by these findings, DFMO@Ui‐Mn is developed as a therapeutic platform combining cascade ROS scavenging with immunometabolic ...
