Arginine is an intermediate in the elimination of excess nitrogen and is the substrate for nitric oxide synthesis. Arginine synthesis has been reported in brain tissue. We have studied the activity of the arginine biosynthetic enzymes argininosuccinate synthetase and argininosuccinate lyase in dexamethasone and/or dibutyryl cyclic AMP treated rat ...

Nitro analogs of substrates for argininosuccinate synthetase and argininosuccinate lyase
Archives of Biochemistry and Biophysics, 1984The nitro analogs of aspartate and argininosuccinate were synthesized and tested as substrates and inhibitors of argininosuccinate synthetase and argininosuccinate lyase, respectively. The Vmax for 3-nitro-2-aminopropionic acid was found to be 60% of the maximal rate of aspartate utilization in the reaction catalyzed by argininosuccinate synthetase ...

Gene complementation analysis of genetically defective cells is a useful tool in the knowledge of the heterogeneity of the inherited diseases. We have used the polyethylene glycol fusion on fibroblasts deficient either in argininosuccinate synthetase (ASS) or in argininosuccinate lyase (ASL) to detect a possible heterogeneity of citrullinemia or ...

Argininosuccinate synthetase and argininosuccinate lyase are soluble cytoplasmic enzymes of the urea cycle. Previous biochemical studies using permeabilized hepatocytes showed that these enzymes are organized in situ, and function as if they are located next to the outer membrane of mitochondria. We have now confirmed and extended those observations in

The Human Argininosuccinate Synthetase Locus and Citrullinemia
1986Argininosuccinate synthetase is an enzyme which functions in the urea cycle in ureotelic animals, and genetic deficiency of the enzyme in man causes an inborn error of metabolism, citrullinemia. The purpose of this chapter is to summarize recent developments in the molecular analysis of the human argininosuccinate synthetase locus and to relate this ...

Advances in Enzyme Regulation, 1980
Abstract Regulation of urea synthesis and the role of ASS were studied in rats subjected to acute dietary transitions from high to low protein or vice versa and in rats injected with an ammonium salt. Significant increase/decrease in urea excretion and urea in the liver preceded increase/decrease of ASS during the dietary transitions.

Postpartum “Psychosis” in Mild Argininosuccinate Synthetase Deficiency
Obstetrics & Gynecology, 2005Urea cycle disorders are relatively rare but well-established causes of postpartum coma and death. Such clinical presentations have been reported previously in ornithine transcarbamylase and carbamyl phosphate synthetase deficiencies. We describe a woman, without prior symptoms of metabolic disease, who presented with hyperammonemia and psychiatric ...

Immunocytochemical localization of argininosuccinate synthetase in the rat brain
Journal of Comparative Neurology, 1991AbstractThe neuronal distribution of argininosuccinate synthetase (ASS) was mapped in the rat brain. Argininosuccinate synthetase is one of the enzymes of the arginine metabolic pathway and catabolizes the synthesis of argininosuccinate from aspartate and citrulline.

Control of argininosuccinate synthetase by arginine in human lymphoblasts
Somatic Cell Genetics, 1978Human lymphoblasts in long-term culture have the enzyme activities necessary to convert citrulline to arginine: argininosuccinate synthetase and argininosuccinate lyase. Upon transfer from arginine-supplemented to citrulline-supplemented medium, lymphoblasts exhibit a lag period before resuming exponential growth.

Qualitative and quantitative abnormalities of argininosuccinate synthetase in citrullinemia
Clinica Chimica Acta, 1981Enzymological and immunochemical analyses of the liver were preformed in seven Japanese patients with citrullinemia. Among the urea cycle enzymes in the liver, only the activity of argininosuccinate synthetase was specifically decreased to 2 to 50% of normal controls.
