Arginine deprivation alters microglial polarity and synergizes with radiation to eradicate non-arginine-auxotrophic glioblastoma tumors

open access: yesThe Journal of Clinical Investigation, 2022
New approaches for the management of glioblastoma (GBM) are an urgent and unmet clinical need. Here, we illustrate that the efficacy of radiotherapy for GBM is strikingly potentiated by concomitant therapy with the arginine-depleting agent ADI-PEG20 in a

Prognostic and Therapeutic Impact of Argininosuccinate Synthetase 1 Control in Bladder Cancer as Monitored Longitudinally by PET Imaging [PDF]

open access: yesCancer Research, 2014
Abstract Targeted therapies have yet to have significant impact on the survival of patients with bladder cancer. In this study, we focused on the urea cycle enzyme argininosuccinate synthetase 1 (ASS1) as a therapeutic target in bladder cancer, based on our discovery of the prognostic and functional import of ASS1 in this setting ...

Case report: Diagnosis of ADCY5-related dyskinesia explaining the entire phenotype in a patient with atypical citrullinemia type I

open access: yesFrontiers in Neurology, 2023
In this case study, we report the case of a 13-year-old girl with citrullinemia type 1 (MIM #215700), an autosomal recessive inherited disorder of the urea cycle, which was confirmed by the identification of a homozygous pathogenic variant in the ...

Rare Adult-onset Citrullinemia Type 1 in the Postpartum Period: A Case Report

open access: yesClinical Practice and Cases in Emergency Medicine, 2023
Introduction: Citrullinemia type 1 (CTLN1) is a urea cycle disorder caused by defective argininosuccinate synthetase leading to impaired ammonia elimination.

Pyrroloquinoline Quinone Attenuates Traumatic Brain Injury-Induced Secondary Damage by Activating PINK1/Parkin-Mediated Mitophagy and Suppressing ASS1/CPS1-Driven Arginine Biosynthesis. [PDF]

open access: yesCNS Neurosci Ther
Pyrroloquinoline quinone (PQQ) attenuates traumatic brain injury in mice. Multi‐omics integration and molecular docking reveal that PQQ activates PINK1/Parkin‐mediated mitophagy to restore mitochondrial homeostasis while suppressing ASS1/CPS1‐driven arginine biosynthesis to dampen neuroinflammation, identifying a dual‐axis therapeutic mechanism for TBI.

Generation of an ASS1 heterozygous knockout human embryonic stem cell line, WAe001-A-13, using CRISPR/Cas9

open access: yesStem Cell Research, 2018
The ASS1 gene encodes argininosuccinate synthetase-1, a cytosolic enzyme with a critical role in the urea cycle. Mutations are found in all ASS1 exons and cause the autosomal recessive disorder citrullinemia. Using CRISPR/Cas9-editing, we established the

Differential expression of argininosuccinate synthetase in serous and non‐serous ovarian carcinomas [PDF]

open access: yes, 2015
The current standard of care for epithelial ovarian cancer does not discriminate between different histologic subtypes (serous, clear cell, endometrioid and mucinous) despite the knowledge that ovarian carcinoma subtypes do not respond uniformly to ...

Defects in intratumoral arginine metabolism attenuate the replication and therapeutic efficacy of oncolytic myxoma virus

open access: yesJournal for ImmunoTherapy of Cancer, 2023
Background Arginine (Arg) is a semiessential amino acid whose bioavailability is required for the in vitro replication of several oncolytic viruses. In vivo, Arg bioavailability is regulated by a combination of dietary intake, protein catabolism, and ...
