Effects of microbial fermentation on enzyme activity and volatile properties of Massa Medicata Fermentata

open access: yesTraditional &Kampo Medicine, Volume 9, Issue 1, Page 10-17, April 2022., 2022
ABSTRACT Aim Massa Medicata Fementata (MMF) is a crude drug used in East Asia to treat anorexia and dyspepsia. It is prepared from wheat and several herbs through microbial fermentation using Aspergillus sp. and Rhizopus sp. There is great difference in the quality of commercial MMF, and the microbes of MMF are suggested to affect its quality.

Taro koji of Amorphophallus konjac enabling hydrolysis of konjac polysaccharides to various biotechnological interest [PDF]

open access: yes, 2020
Due to the indigestibility, utilization of konjac taro, Amorphophallus konjac has been limited only to the Japanese traditional konjac food. Koji preparation with konjac taro was examined to utilize konjac taro as a source of utilizable carbohydrates ...

Effects of Amazake Produced with Different Aspergillus on Gut Barrier and Microbiota

open access: yesFoods, 2023
Inflammatory bowel disease (IBD) is a chronic inflammatory disease of the gastrointestinal tract. To explore the preventive effects of dietary foods on IBD, we evaluated the effects of the traditional Japanese fermented beverage “Amazake” on gut barrier ...

Insights regarding sirtuin-dependent gene regulation during white koji production

open access: yesCommunicative & Integrative Biology, 2022
White koji, a solid-state culture of Aspergillus luchuensis mut. kawachii using grains such as rice and barley, is used as a source of amylolytic enzymes and citric acid for the production of shochu, a traditional Japanese distilled spirit. We previously

A CRISPR/Cas9-mediated gene knockout system inAspergillus luchuensismut.kawachii

open access: yesBioscience, Biotechnology, and Biochemistry, 2020
AbstractWe developed an approach to genome editing of the white koji fungus, Aspergillus luchuensis mut. kawachii using clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 system. Co-transformation of AMA1-based Cas9 and gRNA expression plasmids achieved efficient gene knockout in A. kawachii.

A Strategy for Rapid Acquisition of the β-D-Fructofuranosidase Gene through Chemical Synthesis and New Function of Its Encoded Enzyme to Improve Gel Properties during Yogurt Processing

open access: yesFoods, 2023
A chemical gene synthesis strategy was developed in order to obtain β-D-fructofuranosidase, and a novel gene, AlFFase3, was characterized from Aspergillus luchuensis and expressed in Escherichia coli.

Construction of transcription factor gene deletion library of Aspergillus luchuensis.

open access: yesThe Journal of general and applied microbiology, 2021
We selected 96 genes of Aspergillus luchuensis for the construction of a transcription factor gene deletion library. Of these, we successfully deleted 93 genes using Agrobacterium-mediated transformation (AMT) of A. luchuensis RIB 2604 ΔligD strains.

Development of an itraconazole resistance gene as a dominant selectable marker for transformation in Aspergillus oryzae and Aspergillus luchuensis

open access: yesBioscience, Biotechnology, and Biochemistry, 2021
ABSTRACT There are only a few combinations of antifungal drugs with known resistance marker genes in the Aspergillus species; therefore, the transformation of their wild-type strains is limited. In this study, to develop the novel dominant selectable marker for itraconazole, a fungal cell membrane synthesis inhibitor, we focused on ...
