AMMONIUM OXIDATION BY CELL-FREE EXTRACTS OF ASPERGILLUS WENTII

Cell-free extracts of Aspergillus wentii were found to catalyze the oxidation of ammonium to nitrate; small quantities of hydroxylamine and nitrite accumulated during the oxidation. The oxidations of hydroxylamine to nitrite and of nitrite to nitrate apparently took place through the respective cytochrome c reductases.

Production of amylase by a submerged culture ofAspergillus wentii

Soluble starch was hydrolysed to maltose by Aspergillus wentii Wehmer (IMI 17295). Studies on nutritional requirements of Aspergillus wentii for production of amylase revealed that the optimum conditions were achieved in fermentation culture medium containing 1% starch, and incubated at 20 degrees C for 3 days at pH 6.0.

The production of β-glucosidase in shake-flasks by Aspergillus wentii

Abstract Studies in shake-flasks showed that Aspergillus wentii produces the maximum activity of β-glucosidase among the cultures tested. The activity against cellobiose was about 2–3 fold that against 4NPG. Aspergillus wentii produced a maximum activity of 16.5 U/ml in 14 days on malt extract.

IN the course of an empirical screening programme we found that culture nitrates of Aspergillus wentii, grown on Raulin–Thom medium (5 per cent glucose) supplemented with 0.1 per cent Difco yeast extract, have a pronounced inhibitory effect on the growth of ‘Meteor’ pea seedlings.

Endoglucanase from the mixed culture of Trichoderma reesei and Aspergillus wentii

The level of α-mannanase in mixed fungal culture of Trichoderma reesei, D1-6, and Aspergillus wentii, Pt 2804, affects the extracellular activities of cellulase. The endoglucanase component of the cellulase system is a glycoprotein having mannose and other sugars and sugaramines in its glycan moiety. Its activity is inhibited by α-mannanase.

Ultraviolet induced mutations in Aspergillus wentii.

Aspergillus wentii (IMI 17295) and its three nutritional mutant strains were irradiated with UV rays. New mutants obtained differed from the parent strains in colour of the conidia, growth factor requirements and amylase activity. Arginine deficient strains showed greater amylase activity.
