Cortical‐Hypothalamic Assembloids Uncover the Cortical Regulation of Hypothalamic Responses to Fatty Acid

By assembling cortical‐hypothalamic circuits, we show that cortical input protects hypothalamic neurons from fatty acid‐induced damage, which activates PGC1α to boost mitochondrial function. This reveals a cortico‐hypothalamic axis that defends against metabolic stress.

Degradomics for large‐scale mechanistic insights on proteases and proteolysis in human health

Proteolysis has an important role in human disease but remains relatively unexplored. Degradomics, the uncovering of proteolysis in tissues, cells, and proteins, uses mass spectrometry‐based terminomics to identify protein termini occurring therein (forward degradomics) and to define the actions of proteases (reverse degradomics).

We introduce the MacBrain Resource Center (MBRC) Collections of rhesus macaque embryonic brain tissue. Here, we illustrate histo‐ and immunohistology from our dynamically growing Collection 6, which currently contains >10,000 zoomable and downloadable images from n = 14 male and female embryos from early to late in gestation.

The Isolated Thumb Domain of Acid‐Sensing Ion Channels Forms a Minimal Folding Unit Enabling Ligand Binding Studies

An independently folded thumb domain from acid‐sensing ion channels (ASICs) preserves ligand‐binding and proton‐sensing properties. NMR‐based interaction and pKa analyses uncover how endogenous and exogenous peptides modulate ASICs, providing a simplified model for rational drug discovery targeting proton‐gated ion channels.
