Biofilm Associated Persistence and Drug Tolerance in Mycobacteria Within Host Microenvironments

open access: yesAPMIS, Volume 134, Issue 3, March 2026.
ABSTRACT Biofilms formed by mycobacteria, particularly Mycobacterium tuberculosis (Mtb), represent a major challenge in tuberculosis (TB) treatment due to their highly organized structure and their capacity to induce phenotypic drug tolerance. These three‐dimensional bacterial aggregates are embedded in a self‐produced extracellular matrix that ...

High Prevalence of atpE Mutations in Bedaquiline-Resistant Mycobacterium tuberculosis Isolates, Russia

open access: yesEmerging Infectious Diseases
Bedaquiline is a cornerstone drug for treating drug-resistant tuberculosis. We analyzed 11 isolates from 9 patients who were treated with a bedaquiline-based regimen and remained culture-positive long after treatment start. In 4 of 8 resistant isolates,

Prevalence of Mycobacterium tuberculosis resistant to bedaquiline and delamanid in China

open access: yesJournal of Global Antimicrobial Resistance, 2021
: Objectives: The new antituberculous drugs delamanid and bedaquiline form the last line of defence against drug-resistant tuberculosis (TB). Understanding the background prevalence of resistance to new drugs can help predict the lifetime of these drugs’

Cytotoxicity and Antimicrobial Activity of GaMF1 Analogs

open access: yesChemMedChem, Volume 21, Issue 4, 25 February 2026.
GaMF1 is a reported, highly potent inhibitor of mycobacterial ATP synthase. Structural modification of this scaffold significantly changes biological activity. We present GaMF1 analogues with strong antiparasitic effects, potential antiproliferative properties, and discuss the selectivity limitations of the parent compound and its analogues.

Molecular mechanisms of resistance and treatment efficacy of clofazimine and bedaquiline against Mycobacterium tuberculosis

open access: yesFrontiers in Medicine
Clofazimine (CFZ) and bedaquiline (BDQ) are currently used for the treatment of multidrug-resistant (MDR) Mycobacterium tuberculosis (Mtb) strains.

Bedaquiline, Delamanid, Linezolid, and Clofazimine for Treatment of Pre-extensively Drug-Resistant Tuberculosis

open access: yesClinical Infectious Diseases, 2022
Background Treatment success rates for multidrug-resistant tuberculosis (MDR-TB) remain low globally. Availability of newer drugs has given scope to develop regimens that can be patient-friendly, less toxic, with improved outcomes.

Pharmacokinetic Interactions for Drugs with a Long Half-Life—Evidence for the Need of Model-Based Analysis [PDF]

open access: yes, 2015
Pharmacokinetic drug-drug interactions (DDIs) can lead to undesired drug exposure, resulting in insufficient efficacy or aggravated toxicity. Accurate quantification of DDIs is therefore crucial but may be difficult when full concentration-time profiles ...

Reversing Antibiotic Resistance: Strategies From Adjuvants to Innovative Therapeutics

open access: yesMicrobiologyOpen, Volume 15, Issue 1, February 2026.
Molecular reversal strategies against antibiotic resistance act through three mechanisms: inhibiting resistance gene function, blocking horizontal gene transfer, and modulating host defense. Emerging technologies, such as CRISPR‐Cas gene editing, photodynamic therapy, nanotechnology, and ecological competition, further strengthen these approaches ...

Clinical outcomes of multidrug-resistant tracheobronchial tuberculosis receiving anti-tuberculosis regimens containing bedaquiline or delamanid

open access: yesScientific Reports
The treatment of multidrug-resistant tracheobronchial tuberculosis poses challenges, and research investigating the efficacy of bedaquiline or delamanid as treatment for this condition is limited.
