Solution structure of a bacterial microcompartment targeting peptide and its application in the construction of an ethanol bioreactor [PDF]

open access: yes, 2014
Targeting of proteins to bacterial microcompartments (BMCs) is mediated by an 18-amino-acid peptide sequence. Herein, we report the solution structure of the N-terminal targeting peptide (P18) of PduP, the aldehyde dehydrogenase associated with the 1,2 ...

Mechanistic platform knowledge of concomitant sugar uptake in Escherichia coli BL21(DE3) strains [PDF]

open access: yesScientific Reports, 2017
AbstractWhen producing recombinant proteins, the use of Escherichia coli strain BL21(DE3) in combination with the T7-based pET-expression system is often the method of choice. In a recent study we introduced a mechanistic model describing the correlation of the specific glucose uptake rate (qs,glu) and the corresponding maximum specific lactose uptake ...

PRODUCTION AND CHARACTERIZATION OF CRUDE INTRACELLULER PHYTASE FROM RECOMBINANT BACTERIA pEAS1AMP [PDF]

open access: yes, 2009
This research was aimed at producing a crude intracellular phytase characterized from recombinant bacteria. The recombinant bacteria (pEAS1AMP) was produced by way of transforming pET-22b(+) +pEAS1 into competent E. coli BL21 and E.

\u3cem\u3eArabidopsis thaliana\u3c/em\u3e GLX2-1 Contains a Dinuclear Metal Binding Site, but Is Not a Glyoxalase 2 [PDF]

open access: yes, 2009
In an effort to probe the structure and function of a predicted mitochondrial glyoxalase 2, GLX2-1, from Arabidopsis thaliana, GLX2-1 was cloned, overexpressed, purified and characterized using metal analyses, kinetics, and UV–visible, EPR, and 1H-NMR ...

Cell wall target fragment discovery using a low‐cost, minimal fragment library

open access: yesFEBS Letters, EarlyView.
LoCoFrag100 is a fragment library made up of 100 different compounds. Similarity between the fragments is minimized and 10 different fragments are mixed into a single cocktail, which is soaked to protein crystals. These crystals are analysed by X‐ray crystallography, revealing the binding modes of the bound fragment ligands.

Cloning and expression of native streptokinase in E. coli BL21(DE3)

open access: yesInternational journal of health sciences, 2022
Streptokinase (SK) is a potent plasminogen activator naturally produced by beta-hemolytic streptococcus bacteria. Streptokinase is one of the most important thrombolytic agents that was frequently employed treatment of myocardial infarction. In this study, we amplified the SK gene isolated from Streptococcus pyogenes. The sequence of SK was recorded in

The planar cell polarity protein Vangl2 interacts with the PDZ‐domains of Scribble but not with a unique PDZ‐like domain in Inturned

open access: yesFEBS Letters, EarlyView.
Structural and biochemical characterisations show that the planar cell polarity (PCP) protein Inturned harbours a unique PDZ‐like domain that does not bind canonical PDZ‐binding motifs (PBMs) like that of another PCP protein Vangl2. In contrast, the apical‐basal polarity protein Scribble contains four PDZ domains that bind Vangl2, but one PDZ domain ...

To Construct an Engineered (S)-Equol Resistant E. coli for in Vitro (S)-Equol Production

open access: yesFrontiers in Microbiology, 2018
(S)-equol is one of the major metabolites of daidzein that is produced by human and animal gut bacteria. Most of the physiological functions of soybean isoflavones, such as anti-oxidative activity, anti-cancer activity, and cardiovascular protection have

Identifying Novel Inhibitors of RpFabG in Typhus-inducing Rickettsia prowazekii [PDF]

open access: yes, 2019
Epidemic typhus is a rickettsial disease that is contracted via ticks and lice found on the flying squirrel. The disease is caused by Rickettsia prowazekii, an intracellular, gram-negative coccobacillus.

Generic Model Control Applied to E. coli BL21(DE3) Fed-Batch Cultures [PDF]

open access: yesProcesses, 2020
This work proposes a Generic Model Control (GMC) strategy to regulate biomass growth in fed-batch cultures of Escherichia coli BL21(DE3). The control law is established using a previously validated mechanistic model based on the overflow metabolism paradigm.
